夜交藤提取物对失眠大鼠额叶皮层神经元线粒体结构和功能的影响

摘　　要：【目的】探讨夜交藤提取物对失眠大鼠额叶皮层神经元线粒体超微结构和功能的影响。【方法】选用9月龄SD大鼠,采用对氯苯丙氨酸(PCPA)对大鼠进行造模后,随机分为正常对照组,模型组,安定治疗组(剂量为0.92 mg/kg),褪黑素治疗组(剂量为10 mg/kg)和夜交藤提取物低、中、高剂量治疗组(剂量分别为3、6、12 g/kg);分别采用透射电子显微镜和荧光显微镜观察线粒体的超微结构变化和膜电位改变,并测定线粒体ATP酶含量。【结果】夜交藤提取物治疗后大鼠额叶皮层神经元线粒体超微结构趋于好转;同时随着夜交藤提取物剂量的增加,线粒体Na+-K+-ATPase、Ca2+-Mg2+-ATPase的含量也显著增加(P<0.05)。【结论】夜交藤提取物可能通过改善线粒体的超微结构和增加Na+-K+-ATPase、Ca2+-Mg2+-ATPase的含量而改善失眠。Objective To explore the effect of Ramulus Polygoni Multiflori extracts on mitochondrial uhrastructure and function in the frontal cortex neurons of insomnia rats. Methods Nine-month-old rats were randomly divided into normal control group, model group, antipsychotic treatment group （0.92 mg/kg） , melatonin treatment group （10 mg/kg） and low-, middle-and high-dose Ramulus Polygoni Multiflori extracts treatment groups （3, 6, 12 g/kg, respectively）. Insomnia rat model was induced with para-ehlorophenylalanine （PCPA） . The mitochondrial uhrastructure features and membrane potential changes were observed under electron microscope and fluorescence microscope, respectively. ATP enzyme content in the mitochondria was also determined. Results The mitochondrial uhrastructure pathological changes in the prefrontal cortex neurons of insomnia rats were relieved after treatment with Ramulus Polygoni Muhiflori extracts. With the increase of the dose of Ramulus Polygoni Muhiflori extracts, the contents of Na＋-K＋-ATPase and Ca^2＋-Mg^2＋-ATPase also were increased （P〈0.05） . Conclusion Ramulus Polygoni Multiflori extracts can relieve insomnia probably through improving mitochondrial uhrastructure and through increasing the mitochondrial contents of Na＋-K＋-ATPase and Ca^2＋-Mg^2＋-ATPase.
