地塞米松、氨甲喋呤对人眼球后成纤维细胞增殖的影响

摘　　要：目的　探讨地塞米松、氨甲喋呤对人眼球后成纤维细胞增殖的影响。方法　采用体外培养人眼球后成纤维细胞(HROFb) ,用MTT比色法测定HROFb增殖程度 ,并观察了不同浓度地塞米松、氨甲喋呤单独及联合使用对HROFb生长、增殖的影响。结果和结论　MTT比色法能精确反映HROFb的增殖程度 ,所测定吸收值与细胞数目成正相关 (r =0 .996 ,P <0 .0 0 1) ,地塞米松、氨甲喋呤均可抑制HROFb的增殖 ,且随剂量加大 ,抑制作用增强 ,两者联合使用可减少用量 ,增加抑制作用。Objective:Study on the influences of Dexamethasone and Methotrexate on the proliferation of human retro-ocular fibroblasts(HROFb).Methods:HROFb was cultured and MTT colorimetry assay was used to determine the proliferation of HROFb, the influences of Dexamethasone and methotrexate on the proliferation of HROFb was observed in vitro.Results and Conclusion: MTT colorimetry assay could precisely reflect the proliferation of HROFb. The absorbance was directly proportional to the mumbers of the cells(r=0.996,P<0.001),the proliferation of HROFb was inhibited by Dexamethasone and Methotrexate respectively. The inhibitory effects were stronger when the levels of the agents were higher. Combination of Demethasone and Methoerexate could enhance the inhibitory effects with smaller doses of two agents.
