摘 要:目的探讨二甲双胍对糖尿病大鼠高糖负荷后不同时间点血清胰高血糖素样肽-1(GLP-1)水平的影响。方法选取雌性Wistar大鼠除正常对照组(n=8)外,予以高糖高脂饮食加小剂量链脲佐菌素(25 mg/kg)造模,成模后随机分为模型组(n=8)、二甲双胍组(n=9),分别予以生理盐水、二甲双胍灌胃8 w,作口服葡萄糖耐量(OGTT)试验,并留取各时间点血清用ELISA法检测GLP-1水平。结果与正常对照组比较,模型组大鼠0 h血糖升高(P<0.01),而GLP-1水平较低(P<0.01),高糖负荷后,血糖升高(P<O.01),GLP-1水平亦上升,但仍低于正常对照组(P<0.05);与模型组比较,二甲双胍组大鼠0 h血糖明显降低(P<0.01),高糖负荷后,血糖升高,但明显低于模型组(P<0.05),血清GLP-1水平0 h低于模型组,但高糖负荷后其上升最明显,2 h时最高,但无统计学意义。结论二甲双胍可明显升高高糖负荷后糖尿病大鼠血清GLP-1水平,这可能是其通过促进肠胰岛轴的功能而起到降低血糖的另一机制。Objective To observe the effect of metformin on serum glucagon-like peptide 1 (GLP-1) level in type 2 diabetic rats at different time points after high sugar load.Methods Except the normal control group (n =8),female Wistar rats were chosen to induce diabetes model by high sugar and fat diet and a little dose of STZ (25 mg/kg),then randomly divided into model group (n =8),metformin group (n =9),respectively given saline,metformin for 8 weeks.After the treatment OGTT test was done,serum GLP-1 level at each time point were also tested by ELISA.Results Comnpared with normal control group,the BG level at 0 h of model rats was increased (P < 0.01),while GLP-1 level was decreased (P <0.01),and the BG level at 1 h,2 h after high sugar load significantly were increased,the GLP-1 level also were increased,but still lower than that of the normal control group (P < 0.05).Compared with the model group,the BG level at 0 h of the mnetformin group was decreased (P <0.01),and the BG level at 1 h,2 h after high sugar load were increased,but still lower than that of model group (P <0.05),and the GLP-1 level at 0 h was lower than that of the model group,after high sugar load,the GLP-1 level increased especially at 2 h,but there was no difference statistically.Conclusions Metformin can obviously increase the GLP level of diabetic rats after high sugar load,which may be one of mechanism by promoting enteroinsular axis function.
