摘 要:目的探讨电磁辐射对体外培养的淋巴细胞(Raji细胞)超微结构及其培养上清液中TNF-α水平的影响。方法体外无血清培养Raji细胞,分别用电磁脉冲(EMP)、S波段高功率微波(S-HPM)、X波段高功率微波(X-HPM)为辐射源辐射对数期生长的Raji细胞,采用电镜技术观察辐射6 h时Raji细胞超微结构的改变,各组随机观察100个细胞,通过计数其中受损细胞数对损伤的细胞进行半定量;采用放射免疫分析法检测辐射6 h时Raji细胞培养上清液中TNF-α的水平;所得数据分别采用SPSS 13.0统计学软件的χ2、t检验进行统计学分析,以P<0.05具有统计学意义。结果与对照组相比,各实验组Raji细胞于辐照后6 h超微结构均发生不同程度破坏,损伤的程度以S-HPM最轻,仅表现为线粒体、内质网和核膜结构轻度受损,以X-HPM最重,除细胞器结构严重破坏外,见凋亡小体形成,EMP的损伤程度介于S-HPM和X-HPM之间;半定量结果:与对照组比较,损伤的细胞数S-HPM组最少(P<0.01),X-HPM组最多(P<0.01),EMP组居中间(P<0.01),各实验组间差异显著(P<0.01或0.05)。放射免疫分析法未检测到Raji培养上清液中TNF-α水平的改变(P值均>0.05)。结论 EMP、S-HPM和X-HPM三种电磁辐射6 h均可损伤Raji细胞,导致其超微结构的破坏乃至凋亡坏死,损伤程度与电磁波的种类有关,但不影响Raji细胞培养上清液中TNF-α的水平。Objective To explore the effects of electromagnetic radiation on ultrastructure and TNF-α level in supe in vitro and rnatant of cultured lymphocytes Raji cells. Methods Raji cells were cultured free of serum irradiated in log phase by electromagnetic pulse ( (S-HPM) and X-band high power microwave (X-HPM) , res of Raji cells was observed by electron microscopy and TNF-α EMP), S-band high power microwave peetiv level ely. Six hours later, the uhrastructure in their supernatant was measured by radioimmunoassay. The damaged Raji cells seen under electron microscope were randomly counted out of 100 cells to make a semi-quantitative analysis. Statistical analysis was made by X2 or t test using SPSS 13.0 software. Results The Raji cells chondria, endoplasmic reticulum and nuclear showed uhrastructural changes to a varying extent of mitomembrane at 6 hours after EMP, S-HPM and X-HPM irradiation. No obvious changes were observed in the control effect of S-HPM was slightest, and that of X-I-IPM was Raji cells. Among the three types of radiat most severe with formation of apoptosis, an By th at of EMP was the intermediate. The semi-quantitative analysis showed that the amount of damaged Raji cells was least in the S-HPM group (P 〈 0.01 ), highest in the X-HPM group ( P 〈 0.01 ), and intermediate in the EMP group (P 〈 0.01 ) , and there were significant differences among the three experiment groups (P 〈 0.01 or 0.05 ). TNF-α levels in the Raji cell supernatant of the three groups were not changed significantly assessed by radio immunoassay (P 〉 0.05 ). Conclusions EMP, S-HPM and X-HPM can damage the uhrastructure of Raji cells even induce apoptosis. The extent of damaging effects is related to the types of electromagnetic radiation. However, no obvious effect on the TNF-α level in the Raji cell supernatant is observed.
