摘 要:目的研究蛇床子素对体外培养大鼠股骨组织代谢活性的影响。方法体外分离培养大鼠股骨组织,以雌二醇为阳性对照药物,筛选最佳的药物浓度分别为雌二醇(阳性对照)1×10-8mol/L和蛇床子素1×10-5mol/L,采用终浓度为1×10-5mol/L的蛇床子素对体外培养股骨组织进行药物干预;测定碱性磷酸酶活性、钙含量,实时荧光定量PCR检测骨保护素、核因子κB受体活化因子配体、人类相关转录基因-2以及骨形态发生蛋白-2 mRNA的表达情况。结果 1×10-5mol/L蛇床子素可提高体外培养大鼠股骨组织碱性磷酸酶活性,增加Ca含量,调节骨保护素、核因子κB受体活化因子配体、人类相关转录基因-2和骨形成蛋白-2 mRNA的表达。结论蛇床子素可显著提高体外培养大鼠股骨组织中钙含量、碱性磷酸酶活性以及调节骨代谢相关基因的表达。Objective To investigate the effect of osthole on bone metabolism in rat femoral tissues in vitro. Methods The rat femora[ tissues were isolated in vitro. The optimal concentrations of ostehole ( 1×10^-5mol/L) and estradiol (1×10^-5mol/L) (the positive control) were selected by alkaline phosphatase activity (ALP) . The ALP and calcium levels were detected by commmerical regents, and the expressions of osteoprote- gerin, receptor activator of nuclear factor-KB ligand, runx-related gene 2, and bone morphogenetic protein- 2 mRNA were determined by real-time reverse transcription-polymerase chain reaction. Result The osthole ( 1×10^-5mol/L) significantly increased the activity of ALP, calcium level as well as the expressions of osteoprote- gerin, receptor activator of nuclear factor-KB ligand, runx-related gene-2 and bone morphogenetic protein-2 mR- NA in rat femoral tissues in Vitro. Conclusion Osthole can improve calcium level and ALP activity and the bone metabolism-related genes in rat femoral tissues.
