摘 要:目的:利用血清miRNAs表达谱初步分析荔枝核提取物降低db/db小鼠血糖的可能机制。方法:荔枝核提取物连续灌胃治疗12周,追踪观察其对db/db小鼠体质量、空腹血糖、糖耐量和胰岛素耐量等的影响,并利用血清miRNAs芯片初步分析血清中miRNAs的改变,然后通过Real-time PCR确定组织中相关基因的表达改变。结果:db/db小鼠荔枝核处理组与未处理组相比,体质量、摄食量均无明显改变(P>0.05);但饮水量在灌胃6周时明显减少(P<0.05);空腹血糖值明显下降(P<0.05)。血清miRNAs芯片结果显示db/db小鼠荔枝核处理组miRNAs表达谱出现明显改变,其中均来自于Sfmbt2第10内含子的mmu-miR-297a-3p、mmu-miR-669f-5p、mmu-miR-467d-3p、mmu-miR-466c-5p表达一致性下降;并且Sfmbt2在肝脏组织中表达出现明显下降,为未处理组的64.4%,Sfmbt2在肌肉组织中表达无明显改变。结论:荔枝核灌胃治疗可明显降低db/db小鼠的空腹血糖水平;可改变血清中miRNAs的表达量及肝脏中Sfmbt2的表达量;考虑荔枝核提取物对db/db小鼠的降糖作用可能与miRNAs及Sfmbt2参与的机制有关。Objective:To investigate the mouse glucose decreasing induced by litchi nucleus extracts through serum miRNA expression change as well as the related possible mechanism in db/db mouse model.Methods:After the db/db mouse continuous intragastric administrated with litchi nucleus extracts for 12 weeks,the body weight gain,fasting blood glucose,glucose tolerance and insulin tolerance were all assessed.Plus,miRNAs candidates and their targets genes in serum were also detected and compared with normal control.Results:The body weight gain and the food intake revealed no obvious change in db/db mouse after litchi nucleus extracts treatment compared to normal control (P>0.05),while the amount of water intake at the sixth week of intragastric administration showed decreased significantly (P<0.05); the fasting blood glucose significantly decreased (P<0.05).Serum miRNAs chip data shows that the miRNAs expression changed significantly in db/db mouse from the litchi nucleus extracts treatment group,especially for mmu-miR-297a-3p,mmu-miR-669f-5p,mmu-miR-467d-3p and mmu-miR-466c-5p which all come from Sfmbt2 intron 10.The expression of Sfmbt2 in liver declined significantly,but the expression of Sfmbt2 in muscle tissue showed no obvious change compared to the untreated control group.Conclusion:After continuous intragastric administration with litchi nucleus extracts in the db/db mouse fasting glucose levels changed obviously; expression of serum miRNAs and Sfmbt2 in liver.Thus,we hypothesized that litchi nucleus extracts affect the reduction of fasting blood glucose in db/db mouse via changes of some miRNAs and Sfmbt2 expression profiling.
