摘  要:目的研究不同移植方法对骨骼肌卫星细胞心肌内移植后心肌再生作用的影响。 方法自犬臀部取骨骼肌提取、培养、扩增骨骼肌卫星细胞 ,4’ ,6 -diamidino - 2 - phenylindone(DAPI)荧光标记 ,经局部注射和结扎的左冠状动脉前降支远端灌注移植入自体急性心肌梗塞区域 ,2、4、8周后处死动物 ,取心肌标本行组织形态学检查。结果DAPI标记卫星细胞效果良好 ,在梗塞区、乳头肌及局部注射部位均观察到带荧光的卫星细胞 ,部分已分化成带横纹的肌纤维并以闰盘相连。经冠状动脉灌注移植的卫星细胞排列有序 ,与心肌细胞排列方向一致 ,而局部注射移植的卫星细胞排列紊乱。 结论骨骼肌卫星细胞经冠状动脉灌注移植所形成的肌纤维排列有序 ,分布于整个梗塞区域 ,有利于心肌再生。Objective In this experimental study the effect of different accesses of autologous satellite cell implantation for myocardial regeneration was compared. Methods The satellite cells were procured from the skeletal muscle (gluteus max) of 36 adult mongrel canines, cultured, proliferated and then labeled with 4', 6-diamidino-2-phenylindone(DAPI) in vitro. The cells were autologously implanted into areas of acute left venticular myocardial infarction by local injection or perfusion through the ligated distal left anterior descending coronary artery and the specimens were harvested 2, 4, 8 weeks later for histologic study under both fluorescent microscopy and light microscopy. Results The labeling efficiency of satellite cells with DAPI was almost unexceptional. At 2, 4, 8 weeks, the fluorescent cells were found at the infarcted zone, papillary muscle and local injection site. Some of these cells had progressively differentiated into striated muscle fibers. The satellite cells were implanted by perfusion through the coronary artery and arranged in order of consistency with host myocardial fibers. However, the growing satellite cells, implanted by local injection, were found rather in a mess. Conclusion The satellite cells, implanted by coronary artery perfusion, could progressively differentiate into striated muscle fibers arrange in order, and disseminate all over the infarcted zone in favor of myocardial regene ration .
