摘  要:目的介绍一种快速、有效、高灵敏度的用毛细管气相色谱法测定人血浆中硝苯地平的方法。方法采用SE - 30毛细管色谱柱 (0 .2 4mm× 30m) ,岛津63Ni电子捕获检测器 ,安定作内标。在碱性条件下 ,用乙醚从血浆中提取硝苯地平 ,在气相色谱上直接进样。 结果硝苯地平线性范围为 1~ 10 0 0mg/ml(r =0 .9999) ,最低血浓度检出限 0 .192 pg/ml,绝对回收率 95 %左右。精密度考察硝苯地平日内、日间相对偏差均小于 5 %。硝苯地平在暗室红灯下 ,浓度基本不变。 结论本法适合临床分析研究和应用。Objective A rapid, effective, high-sensitive method for determination of nifedipine in human plasma by capillary gas chromatography is presented. Methods The SE-30 fused-silica column (0.24mm×30m) was used. Diazepam was used as internal standard. Nifedipine was extracted from plasma under basic conditions with ether. Chromatography was performed with an electron capture detector. Results The standard curve was linear in the range 1 ~ 1?000ng/ml (r=0.9999). The minimum detection limit was 0.192pg/ml. The absolute recovery was about 95%. The RSD of intra-day and inter-day assays were less than 5%. The concentration of nifedipine was stable with red light in darkroom. Conclusion It was suitable for clinical research and application.
