摘  要:目的探讨鼻咽癌组织中p14ARF和p27KIP 1基因启动子区甲基化状态在鼻咽癌早期分子诊断和治疗中的作用。方法应用甲基化特异性PCR方法对65例鼻咽癌组织(鼻咽癌组)和30例正常鼻咽上皮组织(对照组)中的p14ARF和p27KIP 1基因启动子区甲基化状态进行检测。结果鼻咽癌组中p14ARF和p27KIP 1基因启动子区甲基化阳性率分别为18.5%(12/65)和61.5%(40/65),两者总检出率为67.7%(44/65);而对照组中均未检测到p14ARF和p27KIP 1基因启动子区甲基化;鼻咽癌组中p14ARF和p27KIP 1基因启动子区甲基化与其临床分期和病理特征均无明显相关性(P>0.05)。同时,p14ARF基因甲基化和p27KIP 1基因甲基化无明显相关性(r=0.05,P>0.05)。结论 p14ARF和p27KIP 1基因甲基化可能是鼻咽癌早期分子事件,可作为鼻咽癌发生的早期诊断指标及甲基化基因治疗的靶点。Objective To study the role of methylation of p14ARF and p27KIP1 genes in the early molecular diagnosis and treatment of nasopharyngeal carcinoma(NPC). Methods p14ARF and p27KIP1 gene methylation status were detected by methylation - specific PCR in 65 cases of NPC tissues ( NPC group) and 30 cases of normal nasopharyngeal epithelial tissues ( control group). Resdts The methylation expression rates of p14ARF and p27KIP1 genes were 18.5% (12/65) and 61.5 % (40/65) respectively in NPC group, while there was no methylation of p14ARF and p27KIP1 genes in control group. The total detection rate of p14ARF and p27KIP1 genes was 67.7% (44/65) in NPC group. The methylation patterns of p14ARF and p27KIP1 genes had no close correlation with the elinieopathologieal features of the NPC patients. In addition, there was no obvious correlation between p14ARF methylation and p27KIP1 methylation ( r = 0.05, P 〉 0.05 ). Conclusion The methylation patterns in p14ARF and p27KIP1 genes may be an early event in NPC, which can be biomarkers for early molecular diagnosis and targets for future genetic therapy of NPC.
