摘 要:目的建立测定右旋糖酐40原料中杂质5-羟甲基糠醛含量的高效液相色谱法。方法采用十八烷基硅烷键合硅胶柱;以1.0g·L-1磷酸溶液-甲醇(90∶10)为流动相;流速为1.0mL·min-1;检测波长为284nm;柱温为(35±5)℃。结果5-羟甲基糠醛在0.10-10.00μg·mL-1质量浓度范围内与峰面积呈良好的线性关系,线性方程为Y=6.7412×104 X-1.1307×103,r=1.000;方法回收率为97.3%,RSD为0.4%。结论该法简便准确,适用于右旋糖酐40原料中5-羟甲基糠醛的含量测定。Objective To establish an HPLC method for the determination of 5-hydroxyme- thylfurfural in the crude material of Dextran 40. Methods The chromatographic conditions adopted were as follows: a C18 column was adopted with a mobile phase of 1.0g . L1 phosphoric acid solution-methanol (90 : 10), with the flow rate being 1.0 mL . min-1 ; the detection wavelength was 284 nm; the column temperature was maintained at (35 ± 5) ~C. Results The content of 5-hydroxyme-thylfurfural had a favorable linear relationship with the peak area within the mass concentration rage of 0.10-10.00.g . mL1, with the linear equation beingY= 6. 7412 X104 X --1.1307 )〈103, r=l. 000; the average recovery was 97.3% and the RSD was 0.4. Conclusion The method is simple and accurate, which is suitable for the determination of 5-hydroxyme-thylfurfural in the crude material of Dextran 40.
