摘  要:【目的】茋类物质是一类重要的次生代谢产物,其在葡萄属植物中承担了重要的病理及生理作用。以往研究发现UV-C照射会引起茋类物质白藜芦醇含量升高,研究UV-C照射后离体叶茋类物质含量与时间的变化关系及与叶龄的关系。【方法】采用不同叶龄的红地球葡萄离体叶片为材料,应用HPLC检测UV-C处理后的不同孵育时间叶片茋类物质的变化规律,以及叶龄对诱导效果的影响特征,同时利用RT-PCR技术对其生物合成途径限速酶基因表达规律进行研究。【结果】UV-C处理后随暗孵育时间的增加,老叶中的茋类物质比率及种类变化较大。诱导后24 h时白藜芦醇苷达到376μg/g,白藜芦醇高达258μg/g,紫檀茋515μg/g,ε-viniferin达到22μg/g,分别是空白组的1.6、50、100以及62倍。【结论】在UV-C处理离体葡萄叶后,叶龄和暗孵育时间对叶片的茋类物质的积累有重要的影响。老叶在UV-C辐射后24 h茋类物质的种类最多,总含量较高。[ Objective] Stilbene belongs to phytoalexin, which plays an important pllyslological a,~u pathological role in grapevines. Previous studies proved the correlation between UV - C treatment and resvera- trol accumulation. However, what remains unclear is the dynamic alteration of stilbenes in response to UV - C treatment on detached leaves differing with ages. [ Method ] The detached leaves of grapevine ( Vitis vinifera L. cv. Red Globe) were employed at two ages whose stilbene species and their contents were assayed with HPLC after UV - C treatments. To evaluate the biosynthetic regulation of those stilbenes, the key enzyme genes in their biosynthetic pathway were semi - quantified by RT - PCR. [ Result ] After UV - C irradiation treatment, the varieties and quantity of stilbenes in leaves during dark duration changed greatly, especially in the elder ones. The contents of stilbenes were raised with 24 h dark duration. Those stilbenes were Pieeid (376μg/g), Resveratrol tents were 1.6, 50, 100, dark duration after UV- C 24 h after induction. (258 μg/g) , Pterostilbene (515μg,/g) , ands- viniferin ( 22 μ g,/g), whose con- and 62 times higher than the control, respectively.
