摘  要:目的:观察益水生新饮对12周龄自发性高血压大鼠(SHR)肾脏组织中TGF-β1、CTGF、FN信号通路的影响,揭示益水生新饮对自发性高血压大鼠肾脏的保护及其作用靶点。方法:将50只12周龄雄性SHR大鼠随机分为:益水生新饮低剂量组[35.55 g/(kg·d)简称低剂量组]、益水生新饮中剂量组[71.10g/(kg·d)简称中剂量组]、益水生新饮高剂量组[142.20g/(kg·d)简称高剂量组]、苯那普利组[10mg/(kg·d)]、模型组,每组10只,模型组生理盐水灌胃,每天1次,共8周。8周后处死大鼠,检测各组大鼠肾组织中TGF—β1、CTGF、FN mRNA和蛋白的检测。观察各组大鼠肾脏病理改变和血压改变。结果:(1)与模型组相比,益水生新饮组和苯那普利组血压明显降低,差异有显著性(P<0.01),其中高剂量组与苯那普利组相比差异有显著性(P<0.01);(2)病理检查提示,与益水生新饮组和苯那普利组相比,模型组系膜细胞及基质增生病变明显加快。(3)与模型组相比,益水生新饮组及苯那普利组肾脏组织中TGF-β1、CTGF及FN表达均下调,差异有显著性(P<0.05,P<0.01),其中,益水生新饮高剂量组与苯那普利组相比,CTGF下调的差异有显著性(P<0.01)。结论:益水生新饮主要通过抑制肾脏中TGF-β1、CTGF及FN信号通路的表达,从而减轻SHR大鼠早期肾脏肾小球硬化和肾间质纤维化的程度,起到延缓高血压的进展和肾脏保护的作用。To study the effect of Yishui Shengxin Decoction on TGF-β1, CTGF and FN signal pathways in kid ney tissue of 12 weeks spontaneously hypertensive rats (SHR). Revealing how did Yishui Shengxin Decoction protect kidney tis sue of SHR and its function targets. Methods: Fifty 12 weeks male SHR were randomly divided into five groups: Yishui Shengxin Decoction low-dose group [35.55 g/ (kg. d), low-dose group for short], Yishui Shengxin Decoction middle-dose group [71. lOg/ (kg. d), middle-dose group for short], Yishui Shengxin Decoction high-dose group [ 14. 20g/ (kg. d), high dose group for short], Benazepril group [ 10mg/ ( kg . d) ], model group, ten for each group, intragastric administration of physiological saline for model group, once a day, eight weeks in all. Put these SHR rats to death after eight weeks. Testing TGF -β1, CTGF, FN mRNA and protein in kidney tissue of each SHR. Observing the changes of renal pathology and blood pressure in each group. Results: (1) Compared with the model group, blood pressure in Yishui Shengxin Decoction group and Benaze pril group had a significant decrease (P〈0.01), compared high-dose group with Benazepril group, there was a significant difference (P〈O. O1 ). (2) Pathological examination showed: compared with Yishui Shengxin Decoction group and Benazepril group, affections of intercapillary ceils and ground substance hyperplasia obviously speeded up in model group. (3) Compared with the model group, expressions of TGF-131, CTGF and FN in kidney tissue of Yishui Shengxin Decoction group and Benaze pril group reduced significantly (P〈0. 05, P〈0. O1 ), compared high-dose group with Benazepril group, down-regulation of CTGF have a significant difference (P〈0.01). Conclusions: Yishui Shengxin Decoction can reduce SHR rats early renal glo- merulosclerosis and renal interstitial fibrosis, mainly through suppression expression of TGF-β1, CTGF and FN signal pathways in kidney, so it plays a role in delaying the prog
