摘 要:以3种中国特产丁香花粉为试材,研究了蔗糖、H3BO3、Ca^2+和GA3对花粉离体萌发的影响,以及贮藏温度和贮藏时间对花粉活力的影响。结果表明:蔗糖、H3BO3、Ca^2+和GA3对3种丁香花粉萌发均有显著影响(P〈0.05)。红丁香、华丁香、白丁香最适宜的花粉离体萌发液体培养基分别是:10%蔗糖+0.01%H3BO3、15%蔗糖+0.01%H3BO3、15%蔗糖+0.05%H3BO3。不同浓度的Ca^2+和GA3对3种丁香花粉萌发的影响有显著差异(P〈0.05),Ca^2+浓度为50~100mg/L和GA3浓度为50mg/L的处理萌发率最高。3种丁香的花粉活力随着贮藏时间的延长逐渐下降,不同贮藏温度对3种丁香的花粉活力影响有显著差异(P〈0.05),-80℃条件下花粉活力下降的速度最慢,是3种丁香花粉最适宜的贮藏温度。The effect of sucrose, H3BO3, Ca^2+ and GA3 on pollen germination in vitro of three special species of Syringa was studied. The influence of storage temperature and time on their pollen viability was also discussed. The results showed that their pollen germination was impacted significantly by sucrose, H3BO3, Ca^2+ and GA3 (P〈0.05). The most suitable pollen germination in vitro culture medium of Syringa villosa, Syringa laciniata, Syringa oblata vat. alba was 10% sucrose + 0.01% H3BO3, 15% sucrose + 0.01% H3BO3, 15% sucrose +0. 05% H3BO3, respectively. The effects of different concentrations of Ca^2+ and GA3 on pollen germination were significantly different(P〈0.05). The germination rate reached the highest when concentrations of Ca^2+ and GA3 was 50-100 mg/L, 50 mg/L, respectively. Along with the extension of storage time, their pollen vitality was declined gradually. The effects of different storage temperatures on pollen vitality were also significantly different (P〈0.05). The pollen vitality declined most slowly under the condition of -80 ℃. Therefore,-80 ℃ is concluded as the most suitable storage temperature for the pollens of the three species of Syringa.
