摘  要:为研究鸭新城疫病毒(Newcastle disease virus,NDV)河北分离株HB/1/05/Dk的遗传变异特性,本试验通过RT—PCR方法扩增其融合蛋白(F)基因片段的5’端的451bp核苷酸,将其推导的氨基酸序列与GenBank已发表的31株NDV参考毒株进行比对分析,并根据F基因(47~420nt)对其进行基因分型。结果表明:鸭NDVHB/1/05/Dk分离株与参考毒株间的核苷酸和氨基酸的同源性分别为61.8%~98.4%和62.9%~97.6%,与鸡源NDVXJ-2/97株同源性最高;HB/1/05/Dk株F蛋白裂解位点氨基酸序列为^112R—R—Q-K—R—F^117,为强毒NDV的特征;该分离株属于基因VIId亚型。表明鸭NDV分离株可能来源于感染NDV的鸡群。Abstract. In order to study the genetic variation of NDV HB/1/05/Dk isolated from Hebei province, the fragment of the fusion protein gene of the strain was amplified by using reverse transcription polymerase chain reaction (RT-PCR) and sequenced. The nucleotide sequence from ATG to 451 bp and deduced amino acid sequence from ATG to 374 bp of F gene of HB/1/ 05/Dk isolate were obtained finally, and analyzed with corresponding sequences of 31 strains of NDV reference published in GenBank. The homology analysis of the nucleotide sequences and amino acids of F gene indicated the isolate have homologies of 61.8%-98.4% and 62.9%- 97.6%/40 with 31 strains, specially the NDV strain XJ-2/97, with 98.4% and 97.6% identity. The amino acid sequence of cleavage site region of HB/1/05/Dk isolate was I-2R-R-Q-K-R-Fn7, which is a typical sequence of velogenic strain. For genotyping, a phylogenetic tree based on nucleotides 1-374 of the F gene was constructed using HB/1/05/Dk and sequences of 31 strains of NDV reference. Phylogenetic analysis showed the isolate was belonged to the genotype VIId virus. The results indicated that chicken flocks may be the source for the NDV isolate HB/1/ 05/Dk.
