摘  要:目的 观察骨髓间充质干细胞(MSCs)对高氧肺损伤新生大鼠TNF-α、IL-10、TGF-β1表达的影响.方法 SD新生大鼠随机分为A、B、C、D、E五组,A、B两组高氧暴露后经尾静脉分别注射1×105/只和1×104/只骨髓MSCs,C组高氧暴露后注射磷酸盐缓冲液(PBS),D、E两组为空气组,分别注射MSCs 1×105/只和PBS,观察MSCs对大鼠肺组织TNF-α、IL-10、TGF-β1表达的影响.结果 C组TNF-α、TGF-β1表达高于E组(P〈0.05);A、B两组治疗后TNF-α及TGF-β1表达降低(P〈0.05),仍高于E组(P〈0.05);C组IL-10表达稍高于E组(P〉0.05);治疗后第3天A、B两组IL-10表达增加(P〈0.05),A组与C组比较差异有统计学意义(P〈0.05),治疗后第14天,各组IL-10表达差异无统计学意义(P〉0.05);D、E两组TNF-α、IL-10、TGF-β1表达差异无统计学意义(P〉0.05).A组对高氧肺损伤的治疗作用优于B组(P〈0.05).结论 两种剂量的MSCs均可以上调高氧肺损伤大鼠肺组织IL-10,降低TNF-α、TGF-β1的表达;高剂量组的治疗效果更明显.Objective To investigate the therapeutic effect of bone marrow mesenchymal stem cells (MSCs) on TNF - a, IL - 10 and TGF - β1 level in hyperoxic lung injury rats. Methods MSCs cultures were prepared from bone marrow of SD rats by adherent culture. Newborn rat pups from four different litters were randomized to A, B, C, D, E groups. Different dose of bone marrow MSCs ( 1 x 105 per pup or 1 × 10^4 per pup) were injected via the tail vein respectively for group A and B after 7 days of oxygen exposure, and phosphate buffered saline (PBS) was injected for group C. MSCs (1×10^5 per pup) and PBS were injected respectively for group D and E as normoxia groups. The lung cytokine level (TNF -α, IL - 10 and TGF - β1 ) were determined. Results TNF -α and TGF - β1 levels were higher in hyperoxia exposed animals (C group) compared with those in normoxia group (E group), P 〈 0.05. MSCs treatment (group A and B) significantly alleviated the elevation of TNF-α and TGF- β1 caused by hyperoxia exposure (TNF -α 107.05±6.43 pg/mL vs. 129.07±6.31 pg/mL, group A vs. group C, P 〈0.05 ; TGF -β1 : 4050.80±474.56 pg/mL vs. 5114.67 ±383.79 pg/mL, group A vs. group C, P 〈0.05). But the TNF- α and TGF- 131 levels in group A and B were still higher than those in group E, P 〈 0.05 . Furthermore, there was no significant difference of IL - 10 in lung between hyperoxia- exposed animals (group C) and normoxia group (group E) , P 〉 0.05. Hyperoxia exposure but MSCs treatment animals (group A and B) presented a significant increase of IL- 10 level compared with the respective PBS controls ( group C ) on post - injection day 3 ( 724. 94 ± 4.84 pg/mL vs. 652.15± 35.99 pg/mL, group A vs. group C, P 〈 O. 05 ) ; But IL - 10 level was not significantly different among the groups on post - injection day 14, P 〉 0.05. There was no significant difference of TNF -α, IL - 10 and TGF -β1 in lung between two normoxia groups ( D and E group), P 〉0.05. The therapeutic
