摘 要:优选基于多孔二氧化硅的肉桂挥发油微球的最佳制备工艺。采用多孔二氧化硅吸附肉桂挥发油后,以海藻酸钠为骨架材料,通过滴制法制备肉桂挥发油微球;以微球粒径及分布、载药量和包封率等作为考察指标,采用L9(34)正交试验对制备工艺进行优化。采用扫描电镜(SEM)、综合热分析(TGA)和红外光谱(IR)等方法对肉桂挥发油微球进行表征,并进行体外释药试验。结果表明按优化工艺参数制备的肉桂挥发油微球形态圆整,粒度均匀,分散性良好,平均粒径为1.61 mm,平均载药量为32.85%,药物包封率为94.79%。在pH 1.0,4.0,6.8,7.4环境中,6 h达到药物最大释放量,分别为50.2%,72.6%,95.0%,97.4%。同时IR,TGA等方法证明了微球的生成。优选制备工艺所得肉桂挥发油微球具有操作稳定性,工艺切实可行。To determine the optimum process for preparing Cinnamomi Cortex oil microspheres based on porous silicon diox- ide. After porous silica dioxide adsorbed Cinnamomi Cortex oil, Cinnamomi Cortex oil microspheres were prepared by the dropping method, with sodium alginate as the skeleton materials. The preparation process was optimized through the L9 (34) orthogonal test de- sign, with microspheres diameter, distribution, drug loading capacity and entrapment efficiency as the indexes. The cinnamon volatile oil microspheres were characterized by scanning election microscope (SEM), thermogravimetrie analysis (TGA), and infrared (IR) spectroscopy. An in vitro drug release experiment was conducted. The resuhs showed that the microspheres prepared with the optimal process parameters were in good shape, even in size and good in dispersibility, with an average diameter of 1.61 mm, an average drug loading capacity of 32. 85% , an entrapment efficiency of 94. 79%. The maximum drug release capacity reached 72. 6% , 95. 0% , 97.4% , respectively, under pH 4. 0, 6. 8, 7.4 in 6 hours. Meanwhile, microsphere generation was tested by IR, TGA and other methods. The established optimum process for preparing Cinnamomi Cortex oil microspheres was proved to be stable and practical.
