摘 要:目的探讨光照对大鼠卵巢功能的影响及滋阴补阳序贯中药对卵巢结构和功能的调节机制。方法采用人工光环境建立大鼠生殖节律失调模型。30只大鼠随机分为空白组、模型组和中药组各10只,中药组采用滋阴方与补阳方序贯给药,给药剂量为1ml/100g,模型组和空白组给予等量生理盐水,各组连续灌胃18d。抽提各组大鼠卵巢组织的总RNA,经逆转录分别用Cy3、Cy5荧光标记,获得各组大鼠卵巢cDNA的探针,再与cDNA基因表达谱芯片杂交,结果由激光扫描仪扫描并进行Ratio值分析、聚类分析。结果聚类分析结果显示,中药组基因谱与模型组的表达差异较大。模型组VS空白组的差异表达基因1 896条,下调1 065条,上调831条;中药组VS模型组的差异表达基因2 582条,下调1 325条,上调1 257条。1 067个差异基因的偏差得到调整,其中611个基因表达增加,456个基因表达下降。差异表达基因的生物学过程结合pathway分析结果表明,中药组对黄体生成素受体、褪黑素受体等通路的基因网络表达调控作用明显。结论滋阴补阳序贯中药可能通过调节细胞增殖、抗凋亡以及甾体激素合成代谢过程等环节部分解除光照致生殖节律紊乱,从而改善卵巢功能。Objective To study the effect and mechanism of sequential yin-nourishing yang-supplementing medicinal on ovarian differential gene expression in rats with reproductive rhythm disorders under artificial light environment. Methods The rat models of reproductive rhythm disorders were established under artificial light environment. Thirty rats were randomized into the blank group, model group and Chinese medicinal group, with 10 in each. The Chinese medicinal group was sequentially administered with 1 ml/100 g yin-nourishing formula and yang-supplementing formula. The model group and blank group were gavaged with same volume of normal saline, The intragastric administration lasted for 18 days in each group. The total RNA in ovarian tissue of rats was extracted with one-step method and fluorescence labeled with Cy3 and Cy5 after reverse transcription. The ovarian cDNA probes of rats in each group were got and then hybridized with eDNA microarray. The results were scanned with laser scanner and the Ratio analysis and duster analysis was conducted with software. Results Cluster analysis showed that the genetic profile in the Chinese medicinal group and model group was quite different. There were 1896 differentially expressed genes (1 065 down-regulating genes and 831 up-regulating genes) as the model group compared with normal group. There were 2 582 differentially expressed genes (1 325 down-regulating genes and 1 257 up-regulating genes) as the Chinese medicinal group compared with model group. The deviation of 1067 differentially expressed genes had been adjusted with increased expression in 611 genes and decreased expression in 456 genes. Differentially expressed genes in biological processes and pathway analysis showed that the regulation of Chinese medicinal on the genetic network expression of luteinizing hormone (LH) receptor and melatonin (MT) receptor was obvious.
