摘  要:以"台农甜蜜桃"胚性愈伤组织为材料,采用RT-PCR和RACE法,首次从台农甜蜜桃中克隆出较为完整的包括Fe-SOD、Mn-SOD、Cu/Zn-SOD 3类型20条的SOD基因家族的全长序列,各成员核酸长度范围在858~1 234bp之间,共编码4种不同的蛋白。生物学分析学分析表明,SOD家族蛋白均为稳定的跨膜蛋白,均含有SOD典型的保守结构域,且在进化上高度保守,但在磷酸化位点上各蛋白差异较大,且3′UTR及5′UTR均具有多态性,可能通过以丝氨酸为主的磷酸化方式,改变酶活性及蛋白构象来参与对桃生长发育的调控。The ‘Premier' Honey Peach EC was used as materials in this experiment.Four different types of 20 mRNA transcripts from SOD gene family,including Fe-SOD,Mn-SOD,Cu/Zn-SOD gene,were cloned by RT-PCR combined with RACE for the first time by reverse transcription polymerase chain reaction(RT-PCR) with rapid amplification of cDNA ends (RACE).The length of nucleic acid of SOD gene family which encoded 4 different proteins were from 858 bp to 1 234 bp.Futher analysis revealed the protein which eneoded from the gene family by bioinformatics had typical conserved domains of SOD and was itransmembrane protein.There were highly conservative in evolution but different SOD proteins had their own respective characteristics of biological property especially on phosphorylation sites.The 3'untranslated regions and 5'untranslated regions of the sequences of SOD gene family had the characteristics of polymorphism polymorphism.The phospho^lation might occur mainly on serine,regulated the growing development of peach by change enzymatic activity and protein conformation.
