摘 要:Rho GAPs(GTPase activating proteins)是真核生物中Rho GTP酶的负调控因子之一,通过激活Rho GTP结合蛋白内在的GTP酶活性,使其加速水解成结合GDP的失活态。全基因组分析指出稻瘟病菌有8个Rho GAP成员(MoRhoGAP),但是其功能不清。本研究利用在线工具分析了其蛋白MoRhoGAP蛋白结构域及其定点突变后的三级结构,以及MoRhoGAP的互作蛋白网络,构建了系统发育树;利用基因芯片和公共数据库分析了在稻瘟病菌不同发育和侵染阶段Rho GAP的基因表达情况,为进一步研究稻瘟病菌RhoGAP的功能提供了理论依据。Rho GAPs(GTPase-activating proteins) are negative regulators of Rho GTPases in eukaryotic organisms.They stimulate the intrinsic Rho GTPase activity and enhance the GTP hydrolysis from the GTP-bound Rho proteins to inactive GDP-bound state,causing their inactivation.Genome annotation shows that the rice blast fungus Magnaporthe oryzae has 8 Rho GAP members,named hereafter as MoRhoGAP.But it is not clear yet of their functions.In this report,we analyzed their functional domains,tertiary structure with site-specific mutations,their interactive proteins and their phylogenetic relationships through bioinformatic methods.Furthermore,we investigated their gene expression profile at different growth and infection stage through DNA-array and public databases.Our study will facilitate the further functional characterization of these RhoGAP members in the fungus.
