摘  要:目的检测类风湿关节炎(RA)及骨关节炎(OA)患者血清中survivin水平,研究其在RA及OA中的变化及其临床相关性。方法用酶联免疫吸附法(ELISA)检测114例RA患者、31例OA患者和30例健康对照者血清的survivin水平,分析血清survivin水平与RA及OA各临床和实验室指标的相关性。结果 (1)RA组患者血清survivin水平高于OA组和对照组(P<0.05);(2)RA患者活动期血清survivin水平与缓解期无统计学差异(P>0.05);(3)RA患者出现X线骨侵蚀血清survivin水平高于非骨侵蚀患者(P<0.01);RA非骨侵蚀患者血清survivin水平高于OA患者(P<0.05);RA骨侵蚀组的病程、类风湿因子(RF)水平及抗环瓜氨酸肽抗体(抗CCP抗体)高于非骨侵蚀组(P<0.05);非骨侵蚀组的病程、RF水平、抗CCP抗体、C反应蛋白及血沉高于OA组(P<0.05);(4)RA组患者血清survivin水平与病程、RF水平、抗CCP抗体、vdH-Sharp评分呈正相关(r分别为0.289、0.385、0.803、0.341,P均<0.01)。结论 RA患者survivin水平增高,并与骨侵蚀性相关,提示其在RA患者的关节损伤持续发展风险方面具有一定意义。Objective To investigate the effects of carbenoxolone on IL-12 and IL-23 expression in EAE mice.Methods The EAE models were established using MOG35-55,and then the mice were intravenously administered with CBX (20 mg/kg) or vehicle control (saline) every other day with the first dose on the day of immunization.The changes of behavioral changes were recorded.Microglia and dendritic cells were incubated with 1μg/mL LPS in the presence of a graded concentration of CBX for 24 h.The IL-12 mRNA and IL-23 mRNA expressions were detected by semiquantitative RT-PCR.Supematants were collected after an additional 24 h culture for ELISA,and IL-12 and IL-23 levels were measured.Results Behavioral changes in CBX group were improved compared with EAE group (P 〈 0.05).CBX negatively regulated the expressions of IL-23 p19 in microglia and dendritic cells.IL-23 production by microglia was decreased in CBX group.Conclusion The level of IL-23 in microglia and dendritic cells was negatively regulated by CBX,and CBX can improve the multiple sclerosis clinical symptoms.
