摘  要:探讨芹菜素对雄性小鼠生殖功能的影响机制,对进一步研究开发芹菜素的药用价值提供一定的线索.将120只6~8周龄的SPF雄性小鼠按体重随机分为4组:对照组、低、中、高剂量组,每组30只.每天定时给予对照组0.01 mL/g bw生理盐水灌胃,实验组给予0.01 mL/g bw不同剂量(低剂量组252mg/L,中剂量组504mg/L,高剂量组1008 mg/L)的芹菜素混悬液灌胃.连续灌胃7、14、21 d后小鼠分批内眦静脉丛取血后处死,测定血清中睾酮(Testosterone,T)和雌二醇(Estradiol,E2);测定精子密度、活动率.血清T浓度14d各剂量组均低于对照组(P=0.000),高剂量组低于中剂量组和低剂量组(P<0.05),小鼠血清T浓度随芹菜素浓度增加出现一定的剂量效应关系.21 d中、低剂量组都低于对照组(P<0.05).21 d低、中、高剂量组的a级精子百分率低于同期阴性对照组(P<0.05).芹菜素对雄性小鼠对精子密度在本实验剂量范围内无明显影响.芹菜素能明显抑制雄性小鼠血清T浓度,进而影响精子的生成.The goal of this study was to investigate the influencing mechanism of apigenin on the reproductive function of male mice.One hundred and twenty of 6 to 8 weeks old SPF male mice were equally randomized into a normal control group and the apigenin groups.Those of the normal control group were given normal saline at a dose of 0.01 mL/g body weight per day.Those of the experimental groups were given different concentration of apigenin (low group:apigenin 252 mg/L,medium group:504 mg/L,and high group:1008 mg/L) at a dose of 0.01 mL/g body weight per day by intragastric infusion.The concentration of serum testosterone (T),estradiol (E2) and sperm motion parameters of those mice were assayed in batches after fed by intragastric administration for 7,14,21 days.The results showed that in 14 d groups,the serum T concentration of each three experimental groups was lower than the normal control group (P =0.000) ;T concentration of the high-dose apigenin group was lower than the medium-dose group and low-dose group (P<0.05).It showed some dose-effect relationship between the T concentration and apigenin.In 21d groups,the T concentration of both medium-dose group and low-dose group were lower than the control (P < 0.05).At 21 days,the percentage of grade a sperm of the mice in each dose group was lower than the control group (P < 0.05).However,no obvious influence on Sperm density of apigenin was detected in this study.These results suggested that apigenin significantly reduced the concentration of testosterone,and hence reduced the production of sperm.
