摘  要:利用HPLC-UV-MS/MS方法对金线莲中黄酮类组分进行分析鉴定和含量测定.液相色谱分离采用ZORBAX Eclipse XDB-C18分析柱,以乙腈和水(含0.03%甲酸)为流动相梯度洗脱,流速为0.5 mL/min.根据ESI-MS/MS分析,并与标准品和文献数据比较,确定金线莲中含有10种黄酮类组分.其中五个已知的黄酮类组分在线性范围内,线性良好(R2≥0.9995),最低检测限(LOD)范围为129.45 ~ 223.56 ng/mL,回收率范围为98~106%(RSD值均小于3.49%).实验表明,所建方法具有高选择性和高灵敏度,适于组培金线莲的质量控制.A HPLC-UV-MS/MS method was developed for simultaneous identification and quantification of flavonoids in Anoectochilus roxburghii (wall.) Lindl (ARL).The chromatographic separation was performed on a ZORBAX Eclipse XDB-C18 column by gradient elution with acetonitrile and water (containing 0.03% formic acid) as mobile phase at a flow rate of 0.5 mL/min.10 compounds were identified or tentatively characterized by online ESI-MS/MS and the comparison with reference compounds and literature data.The limits of detection (LODs) of five known flavonoids were in the range of 129.45-223.56 ng/mL.All calibration curves showed good linear regression (R2 ≥ 0.9995) within the testing concentration ranges.The recoveries were from 98% to 106% with RSD values less than 3.49%.It was demonstrated that the established method was suitable for quality control of ARL,owing to the advantages of enhanced sdectivity and high sensitivity.
