摘  要:目的观察补肾活血通淋方对良性前列腺增生症大鼠模型Bcl-2基因表达的影响。方法 72只Wistar大鼠被随机分为6组,即假手术组、模型组、保列治组(0.8 mg·kg-1)、补肾活血通淋方低、中、高剂量组(7.5,15.0,22.5 g·kg-1)。除假手术组外,余组采用摘除大鼠双侧睾丸后连续注射丙酸睾丸酮(5 mg·kg-1)30 d的方法建立模型,在造模的同时,给药组每天灌胃给药1次,连续30 d。由图象采集和分析系统对每组前列腺组织免疫组化片进行分析,测定Bcl-2表达的阳性平均灰度。结果与模型组比较,各给药组Bcl-2阳性平均灰度均降低(P﹤0.05)。补肾活血通淋方高、中、低剂量组与保列治组比较,Bcl-2阳性平均灰度均降低(P﹤0.05)。结论降低Bcl-2基因的表达可能是补肾活血通淋方治疗良性前列腺增生症的机制之一。OBJECTIVE To observe the effect of Bushen Huoxue Tonglin Formula (BSHXTLF) on gene expression of Bcl-2 in benign prostatic hyperplasia (BPH) rats. METHODS 72 rats were randomly divided into six groups: sham operation group, model group, Proscar group, low dose BSHXTLF group, middle dose BSHXTLF group and high dose BSHXTLF group. The Prosca group was given Prosca at dose of 0.8mg· kg^-1, while BSHXTLF groups were administrated with BSHXTLF at dose of 7.5, 15.0, 22.5g· kg^-1 respectively. The BPH model was established by injecting Testosterone propionate 30 days after Removing Testis in the rats exclude sham operation group. The drugs were administrated once a day for 30 days at the same time. Immunohistochemical film was analyzed by image acquisition and analysis system. RESULTS The positive average gray value of Bcl-2 was decreased in the drug groups compared with model group (P 〈 0.05). The positive average gray values of Bcl-2 were decreased in low dose BSHXTLF group, middle dose BSHXTLF group and high dose BSHXTLF group compared with Proscar group (P 〈 0.05). CONCLUSION BSHXTLF may treat BPH by the mechanism of down-regulated genes expression of Bcl-2.
