摘 要:目的:应用中红外光谱分析技术,建立快速检测牛蒡子中牛蒡苷和牛蒡苷元含量的新方法。方法:用HPLC法测定牛蒡苷和牛蒡苷元的含量,采用偏最小二乘法(PLS),建立中红外光谱与HPLC法测定值之间的多元校正模型,并预测牛蒡子生品和炮制品中牛蒡苷和牛蒡苷元的含量。结果:牛蒡子生品中牛蒡苷和牛蒡苷元的校正模型的相关系数(r)、校正标准偏差(RMSEC)、预测标准偏差(RMSEP)分别为0.998 9,0.068 7,0.951 0和0.999 5,0.022 0,0.322 3;牛蒡子炒制品中牛蒡苷和牛蒡苷元的校正模型的相关系数、校正标准偏差、预测标准偏差分别是0.982 4,0.206 1,1.010和0.995 8,0.069 4,0.406 8。经外部验证,预测值与真实值的相对偏差较小。结论:该方法准确、快速、简便,可实现大批量样品的快速分析。Objective:To develop a mid-infrared quantitative method for rapid determination of arctiin and arctigenin in Fructus Arctii. Methods:Mid-infrared diffuse reflectance spectroscopy technology was used to collect mid-infrared spectra of Fructus Arctii with HPLC analysis values as reference, and the rapid determination method of arctiin and arctigenin was established with partial least squares (PLS). Results:The correlation coefficients (r), root-mean-square error of calibration (RMSEC)and root-mean-square error of validation (RMSEP) of the calibra- tion model for arctiin and arctigenin in Fructus Arctii crude drug were 0. 998 9,0. 068 7,0. 951 0 and 0. 999 5, 0. 022 0,0. 322 3 ,respectively;these parameters of arctiin and arctigenin in Fructus Arctii processed were 0. 982 4, 0. 206 1,1. 010 and 0. 995 8,0. 069 4,0. 406 8 ,respectively. The relative standard deviations(RSDs) of the true value and prediction value from external validation of crude and processed Fructus Arctii were reasonable. Conclusion :The method is accurate,rapid, and simple, and could be applied for the rapid determination of a large number of samples.
