丹栀逍遥散对糖尿病抑郁大鼠肝组织IRS-2—PI3-K信号通路的作用

摘　　要：目的:探讨丹栀逍遥散对糖尿病抑郁大鼠的外周胰岛素抵抗的部分机制。方法:以高脂高糖乳剂加链尿佐菌素复制糖尿病大鼠模型后,再予慢性孤养加慢性不可预见性中等刺激,建立糖尿病抑郁模型,给予丹栀逍遥散5 w后,观察其血糖、血脂、游离脂肪酸、肝糖原、肝组织IRS-2、PI3-K和Akt mRNA的变化。结果:丹栀逍遥散降低糖尿病抑郁大鼠血糖、血脂、游离脂肪酸、升高肝组织IRS-2、PI3-K和Akt mRNA的表达。结论:丹栀逍遥散治疗糖尿病抑郁其机理可能与纠正脂代谢和提高肝组织的IRS-2、PI3-K mRNA基因表达从而改善胰岛素的外周组织胰岛素信号传导有关。Objective：To observe the effects of Danzi Xiaoyao Powder（ DZXYP）, a traditional Chinese compound pre- scription, on IRS - 2--PI3 - K signaling pathway in liver tissues of diabetes mellitus rats with depression and to investi- gate the molecular mechanism of DZXYP in DM with depression. Methods ： Male Sprague - Dawley（SD） rats were admin- istrated with enriched diet and injected with Streptozotocin（ STZ 30 mg/kg） ,then isolated and independent chromic un- predictable mild stimulation（3 W） to build the model of diabetes mellitus rats with depression. The changes of serum fasting blood glucose （FBG） , blood lipids, serum fasting insulin （FINS） , liver glycogen and free fatty acid were observed. The expressions of IRS - 2, PI3 - K mRNA and Akt mRNA in liver tissue were determined with RT - PCR. Results ： Com- pared with the model group, FBG, FINS and blood lipids and free fatty levels in DM rats with depression treated with DZX- YP were decreased significantly （ P 〈 0. 05 ）. HDL - C and liver glycogen level in DZXYP group were increased （ P 〈 . 05 ）. IRS - 2 mRNA and PI3 - K mRNA expressions in DZXYP group were obviously increased as compared with those in the model group （ P 〈 0.05 ）. Conclusion ： The effect of DZXYP in treating DM with depression was probably adjusted the glucose and lipid metabolism and associated with its improvement of IRS -2--PI3K mRNA expressions in liver tissue.
