摘  要:目的探讨C-RP、APN在IR大鼠各组织的表达情况。方法19只SD大鼠,随机分为对照组(NC,n=9)与高脂组(HF,n=10),饲养20周后,行OGTT和胰岛素释放试验(IRT),采用实时荧光定量PCR(RT-PCR)及Northern blot法检测大鼠肝脏、脂肪组织C-RP及APN的表达。结果与NC组比较,HF组血糖、胰岛素水平增加,表明已处于IR状态。Northern blot检测显示,HF组脂肪组织中APN的表达较NC组下降(P<0.01)。RT-PCR显示,HF组肝脏组织C-RP较NC组上升约62%(P<0.01),脂肪组织中C-RP的表达较NC组上升约88%(P<0.01)。结论高脂饲养诱导的IR大鼠,肝脏、脂肪组织C-RP表达增加,而脂肪组织APN表达下降。Objective To explore the expression change of C-reactive protein (C-RP) and adiponectin (APN) in different tissues of insulin resistant rats induced by high fat diet.Methods Nineteen male SD rats were randomized into control group(NC,n=9) and high fat-fed group (HF,n=10).20 weeks later,OGTT and insulin releasing test(IRT) were detected in all groups.The expression of C-RP and APN mRNA in liver and adipose tissue were detected by real time PCR and northern blot analysis respectively.Results Compared with NC group,plasma glucose and insulin of HF group were increased remarkably,which indicated that the high fat-fed rats were in the state of insulin resistance.Compared with NC group,northern bolt analysis showed that the expression of APN mRNA in adipose tissue of HF group was decreased by 64%(P<0.01).Real time PCR results showed that the expression of C-RP mRNA in liver and adipose tissues in HF group was higher than in NC group with 62% and 88% increased(P<0.01).Conclusion The expression of C-RP in liver and adipose tissues of insulin resistant rats induced by high fat diet is increased.However the expression of APN mRNA in adipose tissue is decreased.
