摘 要:目的:观察长期不同负荷运动对大鼠心肌MEK/ERK/p90RSK通路及p38磷酸化水平的影响,探索MAPK对运动诱导的心肌肥大的作用。方法:8周龄雄性SD大鼠经过适应性训练后随机分为9组,安静对照组(C)、中等强度运动即刻组(M0)、运动后6h组(M1)、12h组(M2)、24h组(M3),大强度运动即刻组(H0)、运动后6h组(H1)、12h组(H2)、24h组(H3),每组6只。采用动物跑台进行7周递增负荷训练。观察大鼠心系数变化,在运动后即刻、6h、12h、24h分别测定心肌MEK1/2(Ser217/221)、ERK1/2(Thr202/204)、p90RSK(Thr573/Ser363)、p38(Thr180/Tyr 182)磷酸化。结果:M组和H组心系数显著性高于C组,H组显著性高于M组(P<0.01);运动后中等强度组和大强度组MEK、ERK、p90RSK和p38磷酸化水平不同程度的显著性高于C组,随时间变化逐渐下降至安静水平。结论:1)ERK通路和p38的活性在运动后即刻出现峰值,到24h基本恢复到安静水平;2)与p90RSK相比,MEK和ERK对运动较为敏感;3)ERK通路和p38可能参与了长期运动诱导的心肌肥大,但它们的变化难以判定心肌肥大的性质。Objective:In this study, we focused on the myocardial MAPK pathway, in order to discuss the mechanism of MEK/ERK/p90^RSK pathway and p38 after chronic different intensi- ties exercise. Moreover, we want to dicuss the effects of MAPK on exercise-induced cardic hy- pertrophy. Methods: Adult male SD rats (8-weeks-old) were randomly divided into nine groups after adaptive training: C, M0, M1, M2, M3, H0, H1, H2 and H3, n= 6. Rats took treadmill training of incremental load for 7 weeks. Observed the changes of the heart-to-body weight rations, and phosphorylation level of MEK1/2 (Ser217/221), ERK1/2 (Thr202/ 204) ,p90RsK (Thr573/Ser363) ,p38(Thr180/Tyr182) on time-dependent changes. Results: After 7 weeks' training, the heart-to-body weight rations were significantly increased ( P〈0. 01). The phosphorylation of MEK, ERK, p90^RSK and p38 were increased significantly in both intensity groups, and recovered to normal lever within 24 hs. Conclusion:After chronic trained rats following a single bout exercise, MEK/ERK/p90^RSK pathway and p38 were peaking at im- mediately after exercise, and recovered to normal lever in 24 hs. MEK and ERK were more sensitive to exercise compare to p90^RSK. MEK/ERK/p90^RSK pathway and p38 might precede the development of exercise training-induced cardiac hypertrophy, but their changes can't tell the property of cardiac hypertrophy.
