摘　　要：目的探讨磷酸二酯酶5（Phosphodiesterase 5，PDE5）在大鼠脂肪干细胞（Adipose-Derived Stem Cells，ADSCs）中的表达情况，为PDE5基因siRNA修饰ADSCs移植治疗勃起功能障碍提供理论基础。方法分别采用实时荧光定量PCR和Westem blot技术检测ADSCs和阴茎海绵体平滑肌细胞（Cavernosum Smooth Muscle Cells．CSMCs）中PDE5在mRNA和蛋白水平的表达情况，并采用免疫荧光技术观察PDE5蛋白在ADSCs和CSMCs细胞内的表达与定位。结果PDE5在ADSCs中表达，且均匀分布于细胞浆内，其表达水平显著高于CSMCs（P〈0．05）。结论PDE5在大鼠ADSCs内高表达且定位于细胞浆内，为PDE5基因siRNA修饰ADSCs移植治疗勃起功能障碍的可行性研究奠定了良好的基础。Objective To investigate the expression of phosphodiesterase 5 （PDE5） in rat adipose-derived stem cells （ADSCs）, and establish theoretical foundation for intervention of erectile dysfunction with ADSCs using PDE5 siRNA. Methods The levels of PDE5 mRNA and protein in ADSCs and corpus cavernosum smooth muscle cells （CSMCs） were measured respectively by means of realtime PCR and Western blot, and its expression and localization of PDE5 protein in ADSCs and CSMCs were detected by immunofluorescence. Results PDE5 expressed in ADSCs, and evenly distributed in the cytoplasm, the expression of PDE5 in ADSCs was much higher than that of CSMCs （P〈0.05）. Conclusion The high expression of PDE5 in rat ADSCs an its homogeneous distribution in the cytoplasm, provide a foundation for the intervention therapy of ED with ADSCsusing PDE5 siRNA intervention.
