水通道蛋白AQP5在小鼠骨髓间充质干细胞中的免疫定位

摘　　要：水通道蛋白家族是由能够转运水和小的溶质分子透过细胞膜的膜蛋白分子构成。AQP5属于水选择性通道,其在机体组织内的表达非常广泛。间充质干细胞主要来源于骨髓和脂肪组织,能够诱导分化成间充质组织中的各类细胞。采用PCR、免疫印迹和免疫荧光技术分析了水通道蛋白AQP5在小鼠骨髓间充质干细胞中的表达情况。结果表明:AQP5在RNA和蛋白质水平上都有表达,并且与CD90,CD44在小鼠骨髓间充质干细胞膜上共表达。The aquaporin （AQP） family consists of a number of small integral membrane proteins that transport water and small solutes across the membrane. Recent reports demonstrated that AQP5 is ex- pressed in various cells. However, its expression in mesenchyroal stem cells have not been investigated. Therefore, we examined whether AQP5 could express in mice marrow-derived mesenchymal stem cells by immunofluorescence and immunoblotting analyses. The result show that AQP5 expresses in the membrane of marrow-derived mesenchymal stem cells. Accordingly, AQP5 may play an important role in the differ- entiation of marrow-derived mesenchymal stem cells.
