摘  要:目的 观察大鼠脂肪干细胞在向成骨细胞分化过程中富血小板血浆(PRP)的作用及其与线性高密度聚乙烯(Medpor)支架材料的生物相容性.方法 酶消化法获得脂肪干细胞,将大鼠脂肪干细胞与经PRP孵育的Medpor支架材料复合培养,观察PRP对大鼠脂肪干细胞向成骨细胞分化的影响.结果 酶消化法能成功获得大鼠脂肪干细胞,PRP干预组脂肪基质细胞(ADSCs)的增殖率达33.3%,对照组为8.9%. PRP可以显著增加ADSCs的分化增殖(P<0.05).环境扫描电镜观察,脂肪干细胞能在支架上充分的附着伸展.结论 大鼠脂肪干细胞的成功分离诱导分化,为骨组织工程提供了新的种子细胞来源,PRP是促进该细胞成骨细胞分化的有效的细胞因子组合,Medpor材料可作为骨组织工程的支架材料.Objective To investigate the effect of platelet-rich plasma (PRP) on the diffentiation of rat adipose-derived stem cells (ADSCs) into osteoblasts,and the biocompatibility of ADSCs with a Medpor scaffold.Methods By using enzyme digestion method,ADSCs were obtained,and co-cultured with Medpor scaffold materials incubated with PRP.The effects of PRP on the differentiation of ADSCs into osteoblasts were observed.Results Enzyme digestion method could successfully obtained ADSCs in rats.The ADSCs proliferation rate in PRP intervention group and control group was of 33.3% and 8.9% respectively.PRP could significantly increase the differentiation of ADSCs (P 〈 0.05).ADSCs could adhere to and fully extend on the Medpor scaffold under environmental scanning electron microscope.Conclusion Isolation and differentiation of ADSCs in rats offer a new choice of seed cells for osteal tissue engineering.PRP is an effective cell factor for ADSCs differentiation into osteoblasts.Medpor can be used as a potential scaffold in osteal tissue regeneration.
