通心络胶囊对扩张型心肌病大鼠心功能和心室重构的影响

摘　　要：目的:观察通心络胶囊对扩张型心肌病(DCM)大鼠心功能和心室重构的影响。方法:建立DCM大鼠模型,随机分为B、C、D组,另将同周龄的正常大鼠作为A组。C组和D组分别每日予通心络超微粉0.15 g/kg和1.5 g/kg灌胃,A组和B组给予等体积生理盐水灌胃。4周后,超声心动图测定心功能的变化,左室重量指数(LVMI)和心肌胶原容积分数(CVF)评估心室重构的变化。结果:与A组比较,B、C、D组的左室收缩末期内径(LVESD)和左室舒张末期内径(LVEDD)均明显增加(P<0.01),左室射血分数(LVEF)和左室短轴缩短率(LVFS)均明显降低(P<0.01),LVMI和心肌CVF均明显增加(P<0.01);与B组比较,C组的上述指标均无显著变化(P>0.05),D组的LVEDD无显著变化(P>0.05),而LVESD明显减小(P<0.01),LVEF和LVFS均明显增加(P<0.01),LVMI和心肌CVF均明显下降(P<0.01)。结论:高剂量通心络可以改善扩张型心肌病大鼠的心功能和心室重构。Objective： To explore the effect of tongxinluo capsule on cardiac function and ventricular remodeling in rats with dilated cardiomyopathy （DCM）. Methods=Rats that survived after DCM induction were randomly divided into group B, C and D. Age matched normal rats were used as group A. Rats in group C and D received tongxinluo superfine powder intragastrically at a dosage of 0.15 g/kg and 1.5 g/kg, respectively. Rats in group A and B received the same volume of normal saline as the tongxinluo-treated group. Four weeks later, cardiac function was detected by echocardiography, and ventricular remodeling was assessed by left ventricular mass index （LVMI）and collagen volume fraction （CVF）. Results：Compared with group A, rats in group B, C and D exhibited significantly increased LVEDD, LVESD, LVMI and CVF （P〈0.01）, and significantly decreased LVEF and LVFS （P〈0.01）. However, compared with group B, there was significantly increased LVEF and LVFS （P〈0.01 ）, and decreased LVESD, LVMI and CVF in group D （P〈0.01）. No significant difference in these values was shown between group B and group C（all P〉0.05）. Conclusion： High-dose of tongxinluo effectively improves cardiac function and prevents ventricular remodeling in rats with DCM.
