广州地区幽门螺杆菌儿童分离株空泡毒素基因A的分型

摘　　要：目的探讨广州地区幽门螺杆菌(Hp)儿童分离株空泡毒素基因A(vacA)基因亚型的分布。方法从胃十二指肠疾病患儿胃黏膜标本中分离培养Hp,PCR法测定vacA的m1、m2、s1、s2基因亚型。结果从儿童胃黏膜标本中成功分离出106株Hp,106株中vacA基因m阳性率为94.34%(100/106),阴性率为5.66%(6/106)。m1阳性率为36.00%(36/100),m2阳性率为64.00%(64/100)。s阳性率为96.23%(102/106),阴性率为3.77%(4/106),基因分析显示均为s1。vacA亚型组合s1/m1阳性率为36.73%,s1/m2组合阳性率为63.27%。结论广州地区Hp儿童分离株vacA的优势基因型为vacAs1、vacAm2、vacAs1/m2。Objective To investigate the distribution of vacuolating cytotoxin gene A （vacA） genotypes in Helico- bacterpylori （Hp） isolated from children in Guangzhou. Methods Hp were isolated from gastric mucosa specimens in children with gastroduodenal diseases. PCR was applied for the determination of m1, m2, s1 and s2 genotype of vacA. Results A total of 106 strains Hp were isolated, with vacA m, m1, m2 and s positive rates were 94. 34% （ 100/106）, 36. 00% （36/100）, 64. 00% （64/100） and 96. 23% （102/106）, respectively. The genotype analysis showed that the s genotype was all sl. The positive rates of vacA s1/m1 and s1/m2 combination were 36. 73% and 63.27%, respectively. Conclusion The vacAs1, vacAm2 and vacAs1/m2 are predominant genotypes of Hp strains isolated from children with gastroduodenal diseases in Guangzhou.
