摘 要:目的利用纳米微悬臂梁传感器对李斯特细菌毒力基因溶血素基因(hlyA)进行无标记检测,初步研究这种新型传感技术在基因检测中的应用。方法利用聚合酶链反应技术(PCR)特异性扩增单核增生性李斯特细菌溶血素基因(hlyA),采用自组装单分子膜将DNA探针分子固定在悬臂梁的表面,采用XPS(X射线光电子能谱)进行验证。结果完成表面修饰后,实现了对李斯特细菌毒力基因hlyA基因的无标记检测,最低检测限为0.1μg/mL。结论传感器性能稳定,特异性好,反应迅速。Objective To detect Listeria monocytogenes hlyA gene (without labeling the sam- ple) by mieorcantilever biosensor and to investigate mechanism of the new type biosensor. Methods hlyA DNA was amplified by PCR, the gene probe was immobilized onto the micro- cantilevers by self-assembled monolayers, and a functionalisation method was developed and verified by using XPS. Results After being surface modification, Listeria monoc.ytogenes hlyA DNA was detected by silicon dioxide microcantilevers sensor. The LDL was 0. 1 μg/mL. Con- clusion The biosensor exhibits good performance with high sensitivity, stability and fast re- sponse time.
