超高效液相色谱法测定鸡组织中妥曲珠利及其代谢物的残留量

摘　　要：为建立同时检测鸡肌肉、肝脏和肾脏中妥曲珠利及其代谢物妥曲珠利砜和妥曲珠利亚砜残留量的超高效液相色谱方法，采用乙腈作提取溶剂，过固相萃取小柱净化，超高效液相色谱仪进行检测。结果显示，3种药物在3种组织的线性、精密度、准确性、特异性和稳定性良好。其中，3种药物的回收率均在81．6％～110．0％，日内相对标准偏差均小于19．0，日间相对标准偏差均小于18．3％。妥曲珠利、妥曲珠利砜和妥曲珠利亚砜在肌肉和肝脏中的检测限分别为10、10和15μg／kg，在肾脏中检测限分别为25、25和37．5μg／kg，在3种组织中定量限分别为50、50和75μg／kg，经稳定性试验表明样品稳定、方法可靠。本研究建立了同时检测鸡肌肉、肝脏和肾脏组织中妥曲珠利及其代谢物的超高效液相色谱检测方法，该方法准确可靠，重现性好，完全能满足残留检测的要求。To establish the Ultra-high performance liquid chromatography （UPLC）method to simultaneously determine the residues of tohrazuril and its metabolites toltrazuril sulphone and toltrazuril sulphoxide in chicken tissues, the analytes were extracted by aeetonitrile and then the extract was purified by Oasis MAX SPE cartridges, after being concentrated and filtered through 0.22 μm membrane and direetlyinjected into the UPLC system. The calibration curves were linear （R2〉0.99） in the range of 25 to 1 000 μg/kg fortoltrazuril and toltrazuril sulphone and 37.5 to 1 500μg/kg for toltrazuril sulphoxide. The accuracy values lay between 70% and l l0% and the precision was lower than 19.0%. Toltrazuril and tohrazuril sulphone＇s limits of detection and quantitation were 10 and 50 μg/kg in muscle and liver and 25 and 50 μg/kg in kidney, respectively. Toltrazuril sulphoxide＇s limits of detection and quantitation were 15 and 75 μg/kg in muscle and liver and 37.5 and 75 μg/kg in kidney,respectively.
