摘 要:采用HPLC法测定恩诺沙星可溶性粉的含量,色谱柱选用Waters Symmetry C18柱(4.6×150mm,5μm),流动相为0.025 mol/L磷酸(用三乙胺调节pH至3.0)—乙腈(83∶17),检测波长为278 nm,柱温25℃,流速1.0 mL/min.结果表明恩诺沙星在20~80 μg/mL的范围内,呈现良好的线性关系,平均回收率为99.3%.通过试验确定本法的检测限为15 μg/mL,定量限为50 μg/mL,且具有良好的专属性和耐用性.运用本法对来自四个不同厂家的恩诺沙星可溶性粉进行含量测定,并与紫外分光光度法测定结果比较,说明HPLC法能准确测定恩诺沙星可溶性粉的含量.A HPLC method for the determination of enrofloxacin soluble powder has been used on Waters Symmetry C18 column(4.6 × 150 mm,5 μm),with 0.025 mol/L phosphoric acid (adjust pH to 3.0 by triethylamine)-acetonitrile (83 ∶ 17) as mobile phase,UV detection at 278 nm,the column temperature was 25 ℃ and flow rate was 1.0 mL/min.The method showed good specificity,durability and linear relationship within the range of 20 ~ 80μg/mL.LOD and LOQ of enrofloxacin were determined as 15ng/mL and 50 ng/mL.The average recovery of enrofloxacin was 99.3 %.Determination of enrofloxacin soluble powder from four different manufacturers by HPLC and UV spectrophotometry indicated that the HPLC method can accurately control the quality of enrofloxacin soluble powder.
