摘 要:目的:建立同时测定红花注射液中腺苷和羟基红花黄色素A含量的方法。方法:采用高效液相色谱法。色谱柱为AgilentEclipSeXDB—C18(250mm×4.6mm,5μm),流动相为甲醇一乙腈.0.7%磷酸溶液(梯度洗脱),柱温为30℃,流速为0.8ml/min,检测波长为260nm(0~14min)和403nm(〉14~35min)。结果:腺苷和羟基红花黄色素A的质量浓度分别在1.194~71.640、2.070~82.800gg/ml范围内与各自峰面积积分值呈良好的线性关系(r均为0.9999);精密度、重复性、稳定性试验的RSD〈1%;平均加样回收率分别为101.18%和100.56%,RSD分别为1.33%和1.13%(H均为9)。结论:该方法简便、准确、重复性好,可用于红花注射液的质量控制。OBJECTIVE: To develop a method for the content determination of adenosine and hydroxysafflor yellow A in Hong- hua injection. METHODS: HPLC method was adopted. The determination was performed on Agilent Eclipse XDB-C18(250 mm× 4.6 mm, 5 μm)with mobile phase composed of methanol-acetonitrile-0.7%phosphoric acid (gradient elution) at the flow rate of 0.8 ml/min. The column temperature was 30 ℃. The detection wavelength was set at 260 nm within 0-14 min and 403 nm within 14-35 min. RESULTS: The linear ranges of adenosine and hydroxysafflor yellow A were 1.194-71.640 μg/ml(r=0.999 9) and 2.070-82.800 μg/ml(r=0.999 9). RSDs of precision, reproducibility and stability tests were all lower than 1%. The average recov- eries were 101.18% (RSD=1.33% ,n=9) and 100.56% (RSD=1.13% ,n=9). CONCLUSIONS: The methods is simple, accurate and repeatable, and it is suitable for the guality control of Honghua injections.
