摘 要:应用荧光AFLP和拮抗实验对26株灵芝菌种进行了遗传多样性分析。荧光AFLP分析的结果表明,采用9对EcoRI/MseI引物(其中MseI引物为FAM荧光标记物)对26个菌株进行AFLP扩增,共得到1944条清晰易辨的多态性条带。聚类分析表明,26株灵芝菌株在遗传相似系数0.28处聚为3类,分别是美国灵芝类、赤芝类和树舌类。种间的遗传相似系数变化范围在0.23—0.73。荧光AFLP分析结果与拮抗性实验结果基本一致,可见拮抗试验在灵芝亲缘关系的初步鉴定中是有效的,荧光AFLP则更能区分出亲缘关系较近的灵芝菌株。Genetic diversity between 26 strains of Ganoderma was analyzed by fluorescent-AFLP and antagonistic reaction. The dendrogram derived from fluorescent-AFLP showed that 1 944 AFLP polymorphic bands were detected by 9 pairs of EcoR I/Mse I ( MseI-a FAM fluorescent marked primer). UPGM results indicated the genetic relationships of 26 Ganoderma lucidum strains had diversity and similarity. They were classified into 3 categories in 0. 28 coefficients, which was Meiguolingzhi, Chizhi and Shushelingzhi respectively. Genetic similarity coefficient of 26 Ganoderma lucidum strains was in the range of 0. 23 to 0. 73. The results of fluorescent-AFLP were nearly in accordance with that of antagonistic reaction, which showed that the method of antagonistic reaction was effective in the preliminary identification of Ganoderma lucidum strains, and the fluorescent-AFLP was more effective to distinguish the species with more nearly affinity.
