摘 要:采用荧光光谱法和紫外光谱法比较研究了新型含空间链四-(6-氨基己酸磺肽基)铝氯酞菁(AlPc(SO2NHC5H10COOH)4)及AlPc(SO2NHC5H10COOH)4/牛血清白蛋白(BSA)复合物的光物理性质。与无空间链四羧基铝氯酞菁(AlPc(COOH)4)相比较,AlPc(SO2NHC5H10COOH)4不易聚集,荧光强度增强,荧光寿命增长。当AlPc(SO2NHC5H10COOH)4和AlPc(COOH)4分别与BSA形成复合物后,与相应的自由酞菁相比,AlPc(COOH)4/BSA和AlPc(SO2-NHC5H10COOH)4/BSA复合物的紫外光谱红移,荧光光谱猝灭,其荧光猝灭原因为静态猝灭。由于空间链效应,AlPc(SO2NHC5H10COOH)4与BSA之间的作用明显强于AlPc(COOH)4与BSA之间的作用。The purpose of this study was to investigate the photophysical properties of 3rd generation photosensi- tizer using in photodynamic therapy. The photophysical properties of tetra-carboxyl-aminohexanoic-sulfonyl phthalocyanine aluminium chloride (A1Pc (SO2 N HC2 H10 CO0 H) 4 )-bovine serum albumin (BSA) bioconj ugate was studied by UV-Vis and fluorescence spectroscopic method. The results indicated A1Pc (SO2 NHC5 H10COOH)4 exhibited lower aggregation, higher fluorescence intensity and longer lifetime compared with tetra-carboxyl phthaloeyanine aluminium chloride (A1Pc(COOH)4) which may due to the spacer hindrance effect. When A1Pc(SO2NHC4H10COOH)4 and A1Pc(COOH)4 conjugates with BSA, compared with the corre- sponding free phthalocyanine, the UV-Vis spectra of A1Pc(COOH)4/BSA and A1Pc(SO2NHCsH10COOH)4/ BSA was red shift and fluorescence spectra was quenched. Furthermore, A1Pc(SO2NHCsHloCOOH)4 conju- gate also showed stronger combining interaction with bovine serum albumin than that of A1Pc(COOH)4 due to the spacer hindrance effect.
