摘 要:目的 考察含药血清方法的可靠性。方法 自 1d龄 SD大鼠四肢长骨获得破骨细胞 ,接种于象牙薄片上 ,于培养第 7d取出 ,计数各组骨吸收陷窝的数量。选用具有明显抑制破骨细胞骨吸收活性作用的西药福善美、固邦和倍美力 ,以等效剂量给大鼠灌胃后制备含药血清 ,分别与相应药物直接添加的 3个浓度 (1× 10 - 8M、 1× 10 - 6 M、 1× 10 - 4 M)组进行比较 ,观察骨吸收陷窝数量的变化。同等条件下制备生理盐水灌胃对照血清。结果 3种药物含药血清及其直接添加各组的骨吸收陷窝数量均显著性地低于对照组。其中 ,福善美含药血清组与直接添加的 3个浓度组之间无显著性差异 (P>0 .0 5 ) ,固邦和倍美力含药血清组与 10 - 8M直接添加组之间无显著性差异 (P>0 .0 5 ) ,但作用强度不及 10 - 6 M和 10 - 4 M直接添加组 (P<0 .0 5或 P<0 .0 1)。结论 含药血清方法具有一定的可靠性 ,本实验条件下制备的含药血清 ,其药效强度大致相当于直接添加的 10 - 8M浓度。Objective Observe the reliability of the medicinal serum method.Method Get osteoclasts from one-day-old SD rats' long bones,vaccinate them on thin ivory slices,get out at the 7th day of culture,count the bone absorptive lacunae in each group.Choose western medicines Fushanmei,Guban and Beimeili which can apparently inhibit the bone absorptive action of osteoclasts,make medicinal serum by stomach-perfusion with equivalent dosages which are respectively compared with the groups direct added with corresponding medicines in three concentrations(1×10 -8 M,1×10 -6 M,1×10 -4 M),then observe the changes of bone absorptive lacunae.Under the same condition make control group with stomach-perfusion of saline and get its serum.Result The medicinal serum of three medicines and the lacunae of each group with direct addition are much lower than that of control.Among which the medicinal serum group of Fushanmei and three concentrations groups of direct addition have no marked difference(P>0 05),so do medicinal serum group of Guban and Beimeili with direct addition of 10 -8 M,but the their acting strength is lower than that of direct addition group of 10 -8 M and 10 -4 M(P<0 05 or P<0 01).Conclusion The medicinal serum method has a somewhat reliability,the pharmacological intensity of the medicinal serum made under this experimental condition roughly equals to that of direct addition in concentration 10 -8 M.
