摘 要:为研究乙型脑炎病毒(JEV)NS3蛋白对其体外增殖能力的影响,为JEV疫苗的大量生产提供一定的理论基础,构建真核重组质粒pCI-neo-NS3并转染至BHK-21细胞内,接种JEV后于不同时间点采集细胞内以及细胞外病毒,采用荧光定量RT-PCR法检测各时间点细胞内和细胞外病毒含量,同时设立不同质量浓度NS3原核表达蛋白对JEV复制影响对照组。用SPSS 21.0统计分析软件分析,比较病毒增殖情况。分析结果显示转染含NS3基因的真核载体组细胞内和上清中病毒含量都极显著高于对照组(P<0.01),加入NS3原核表达蛋白组上清中病毒含量极显著低于对照组(P<0.01)。To study the impact of Japanese encephalitis virus(JEV) NS3 protein on virus's prolifera- tive capacity in vitro and to provide a theoretical basis for the development of JEV vaccines,an eukaryotic recombinant plasmid pCI-neo-NS3 was constructed and transfected into BHK-21 cells. After inoculated with JEV,the intracellular and extracellular virus was collected at different time points, and JEV content was detected by real-time RT-PCR. The influence of prokaryotic expressed NS3 protein on JEV replication was studied,and the data were analyzed using SPSS 21.0 to compare the proliferation of JEV. Statistical analysis showed that the virus content of intracellular and supernatant in the infected group was signifi- cantly higher than that of the control groups(P〈0.01). However, the virus content of supernatant in the group which was added with the prokaryotic expressed NS3 protein was significantly lower than that of the control groups(P〈0.01). This study confirmed that the NS3 protein has obviously effect on replication of JEV.
