摘 要:目的:探讨宁夏回、汉族人群脂联素基因SNP+276G/T单核苷酸多态性与冠心病的相关性。方法:收集回、汉族冠心病患者95例和93例以及回、汉族健康体检者(对照组)98例和102例,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测SNP+276G/T单核苷酸多态性,酶联免疫吸附法(ELISA)检测血清脂联素浓度。结果:回、汉族人群脂联素基因SNP+276G/T多态GG、TG、TT基因型分布频率冠心病组分别为0.137、0.789、0.074和0.581、0.355、0.064;对照组分别为0.316、0.531、0.153和0.598、0.373、0.029。G及T等位基因分布频率冠心病组分别为0.532、0.468和0.242、0.758;对照组分别为0.582、0.418和0.216、0.784。脂联素基因SNP+276G/T多态GG,TG,TT基因型分布频率在回族冠心病组及对照组间差异有统计学意义(P<0.05);G及T等位基因分布在2个民族冠心病组与对照组比较差异均无统计学意义(P>0.05)。宁夏回、汉族冠心病组血清脂联素水平均低于对照组(P<0.05),回、汉族人群TT+TG基因型血清脂联素水平与GG型相比均无显著差异(P>0.05)。Logistic回归分析结果显示,回族人群中携带TT+TG基因型者发生冠心病的风险增加(OR=5.814,95%CI:3.332~10.144)。结论:脂联素基因SNP+276G/T多态性可能与宁夏回族冠心病相关,与宁夏汉族冠心病无关。Objective:To investigate relationship between adiponectin gene +276 nucleotide G/T polymorphism with coronary heart disease(CHD)in Hui and Han nationalities in Ningxia.Method:The study was performed in 95 Hui patients and 93 Han patients with CHD and was performed in 98 Hui healthy peoples and 102 Han healthy peoples(NC group).Polymerase chain reaction-restriction fragment length polymorphism(PCRRFLP)and enzyme-linked immunosorbent assay(ELISA)were used to detect the gene polymorphism of adiponectin and the serum adiponectin level.Result:The frequencies of 276G/T genotypes of GG,TG,and TT were 0.137,0.789 and 0.074in Hui CHD patients and 0.316,0.531and 0.153in NC group;0.581,0.355and 0.064in Han CHD patients and 0.598,0.373and 0.029 in NC group respectively.The frequencies of G and T were 0.532and0.468,and 0.242and 0.758in CHD group;0.582and 0.418,and 0.216 and 0.784in NC group.There was significant difference in genotypes between CHD group and NC group in Hui nationality(P0.05).There was no significant difference in allele frequencies in Hui and Han nationalities(P0.05).Compared with the NC group,the serum adiponectin level of CHD group was lower than those of NC group in both Hui and Han nationalities(P0.05).No matter Hui or Han nationality,there was no significant difference of serum adiponectin level between TG+GG genotype and TT genotype(P0.05).Logistic regression analysis showed that those carrying TT+TG genotype had an increased risk of CHD in Hui population(OR=5.814,95%CI:3.332-10.144).Conclusion:+276 nucleotide G/T polymorphism of adiponectin gene might be associated with CHD in Hui population but not in Han population in Ningxia.
