摘 要:目的:建立测定人血浆中丙泊酚浓度的RP-HPLC法.方法:以麝香草酚(thymol)为内标,血浆中加入缓冲液(0.1 mol/L NaH2PO4)混匀后经环己烷萃取,吸取有机相,加入四甲基氢氧化铵-异丙醇稀释液,氮气吹干,以流动相甲醇-水(80∶20)溶解.色谱柱:Diamonsil C18柱(200 mm×4.6 mm,5μm);流速:1 ml/min;检测波长:270 nm;柱温:30℃.结果:丙泊酚血药浓度在0.20~~ 10.00 μg/ml范围内线性关系良好(r=0.9997),最低定量限为0.20 μg/ml.低、中、高3种浓度的方法回收率为98.37%~100.26%,提取回收率为83.47%~92.11%.日内RSD为4.72%~9.70%,日间RSD为4.27%~6.41%.结论:该方法适用于丙泊酚的人体药动学研究和血药浓度检测.Objective:To establish a RP-HPLC method for the determination of propofol concentration in human plasma.Methods:Thymol was used as internal standard,plasma was extracted with cyclohexane after it was added into the buffer solution (0.1 mol/L NaH2 PO4).The organic phase was added into tetramethyl ammonium hydroxide and 2-propanol diluted solution,then was dried under the stream of nitrogen.The residue was dissolved by mobile phase of methanol-water (80:20).Separation was performed on a Diamonsil C18 column (200 mm × 4.6 mm,5 μm) and the mobile phase was methanol-water(80 ∶ 20) with the flow rate of 1 ml/min.The column temperature was 30 C and detection wavelength was 270 nm.Results:Propofol presented a good linearity within the range of 0.20-10.00 μg/ml (r=0.999 7).The lowest limit of quantitation was 0.20 pg/ml.The recovery rates of low,medium and high concentrations were 98.37 %-100.26 % and extractive recovery rates were 83.47%-92.11%.It had good intra-day and inter-day precision.The RSD of intra-day was 4.72%-9.70% and inter-day was 4.27%-6.41%.Conclusion:This method can be used in pharmacokinetic study and plasma level determination of propofol.
