摘  要:为了构建14份国产甜菜品种的指纹图谱,利用6个差异较大的甜菜品系对50对SSR引物进行扩增,用8%的非变性聚丙烯酰胺凝胶对SSR-PCR产物进行分离,快速银染法进行染色,共选出7对多态性高、带型清晰、重复性好的SSR引物。利用这7对SSR引物对国产14份糖甜菜品种进行扩增,共获得40条谱带,其中多态性位点28个,多态性比率达75%;每对引物产生的等位基因数为5~8个,平均6.6个。利用其中3对引物S6、S13和BVGTT6构建了14份国产甜菜品种的DNA指纹图谱,结果表明,每个甜菜品种有唯一的数字指纹区别于其他品种,说明SSR标记适用于甜菜品种纯度和真实性的鉴定,同时甜菜品种指纹图谱库的构建也为将来可能出现的甜菜品种纠纷打下坚实的技术基础。To build the finger-print of 4 domestic beet varieties, six beet series which are different from each other magnificently were used to screen 50 couples of SSR primer, SSR-PCR products were separated by 8%non-denaturing polyacrylamide gel, fast silver staining dyed, and at last picked out 7 couples of SSR primer with high polymorphism, clear bands and good repeatability. The 7 couples of SSR were made to expand the domestic 14 beet varieties and altogether got 40 bands, among which polymorphism locus were 28%or 75%of the total number. Alleles generated from each couple of primer were from 5 to 8, with the average number standing at 6.6. Three couples of primes (S6, S13 and BVGTT6) were utilized to build 14 DNA finger-print of domestic beet varieties. The result indicated that each beet variety had a unique digital finger-print which was different from others. It meant that SSR was applicable for identifying the purity and realness of the beet variety. Besides, the development of the finger-print bank database of beet varieties also laid a solid technological foundation for the settlement of the beet variety disputes in the future.
