摘  要:本研究主要揭示炎热气候条件下母猪妊娠后期各组织器官GRαmRNA的表达规律,以期为GRα在母猪的热应激环境下的分子生理机制研究提供参考。试验选用12头妊娠母猪,根据妊娠日龄不同分为2组,每组6头,分别是妊娠90、110天。按照常规方法屠宰母猪,迅速采集脑、卵巢、心脏、肝脏、脾脏、肺脏和肾脏,液氮速冻后-80℃保存,提取RNA,反转录,利用实时荧光定量PCR(qRT-PCR)方法检测各组织中GRαmRNA的相对表达量。结果显示:妊娠90天和110天时,GRα的mRNA表达丰度依次分别表现为:90天丰度,肺脏>脾脏>肝脏>肾脏>脑>卵巢>心脏;110天丰度,脾脏>肺脏>肝脏>肾脏>卵巢>心脏>脑。其次,妊娠110天与90天相比,GRαmRNA的相对表达量除了在肺脏和脑中极显著降低(P<0.01),在肝脏中显著降低之外(P<0.05),其他组织则未表现出显著性变化(P>0.05)。结果表明:炎热气候条件下,母猪体内GRαmRNA优先在肺脏和脾脏中表达,其次是肝脏和肾脏,在脑、卵巢和心脏中的表达量最低,这种变化随妊娠时间延长更为明显,但同时表现出较明显的组织特异性。The study was conducted to reveal the GRα mRNA expression in different tissues of gilts during late gestation in high ambient temperature.The object of this study was to investigate the relationship between the GRα mRNA expression and physiological development of gilts during late gestation,to provide the basic reference data for the study of molecular mechanisms of heat stress.Twelve gilts(Landrace × Large White) during late gestation were assigned into two treatments(6 gilts/treatment) according to the gestational day(90 d and 110 d).The gilts were slaughtered and tissues including brain,ovary,heart,liver,spleen,lung and kidney were collected aseptically over liquid nitrogen and frozen at-80℃.Then total RNA isolation and reverse transcription,quantitative Real-time PCR(qRT-PCR) was performed to determine the mRNA expression of GRα in different tissues of gilts during late gestation. The results showed that the mRNA expression level of GRα in different tissues of gilts at 90 d of gestation was as follows: lung&gt;spleen&gt;liver&gt;kidney&gt;brain&gt;ovary&gt;heart; 110 d: spleen&gt;lung&gt;liver&gt;kidney&gt;ovary&gt;heart&gt;brain. Compared 110 d with 90 d of gestation, except that the expression of GRα in lung and brain was significantly reduced(P&lt;0.01), and was reduced(P&lt;0.05) in liver,there were no gestation stage dependent effects on GRα mRNA expression in all the other tissues(P&gt;0.05). The results indicated that the mRNA expression of GRα was higher in lung and spleen of gilts during late gestation,to a lesser degree in liver and kidney, lower expression levels of GRα were observed in brain, ovary and heart.Compared 110 d with 90 d of gestation, the expression of GRα mRNA were reduced in lung, brain and liver at different degrees. The changes with prolonged pregnancy was more obvious, but at the same time showed a distinct tissue specificity.
