摘 要:目的建立甘草次酸-丹参酮ⅡA复方脂质体(GT-Lip)包封率的测定方法。方法分别采用葡聚糖凝胶过滤法、低速离心法分离脂质体和游离药物。以HPLC法为分析手段,比较该2种方法包封率的测定结果。结果葡聚糖凝胶G-50(M)对脂质体有一定吸附,不适合GT-Lip包封率的测定;低速离心法能使GT-Lip脂质体与游离药物有效分离,经HPLC法测定,丹参酮ⅡA(TSN)与甘草次酸(GA)平均包封率分别达到(81.50±0.76)%和(98.63±0.90)%。结论 HPLC法与低速离心法相结合可快速、有效用于甘草次酸-丹参酮ⅡA复方脂质体的包封率测定。Objective To develop a method for determining the entrapment efficiency of glycyrrhetinic acid-tanshinone IIA compound liposomes (GT-Lip).Methods Sephadex filtration and low-speed centrifugation were used for separating GT-Lip and free drugs.The determination results of these two methods for the entrapment efficiency were compared taken HPLC as analysis approach.Results Sephadex filtration G-50 (M) absorbed some GT-Lip,which was not good for the determination of the entrapment efficiency of GT-Lip.The low-speed centrifugation separated effectively GT-Lip and free drugs,and the average entrapment efficiency of tanshinone IIA reached to (81.50 ± 0.76) % and that of glycyrrhetinic acid reached to (98.63 ± 0.90) % after HPLC determination.Conclusion HPLC combining low-speed centrifugation can applied for the effective determination of the entrapment efficiency of GT-Lip,which is quick and accurate.
