摘  要:目的 建立测定五味沙棘颗粒中异鼠李素含量的高效液相色谱法.方法 采用Kromsail C18柱(250 mm×4.6 mm,5μm),四氢呋喃-乙腈-0.4%磷酸水溶液(12:18:70)为流动相,进行等度洗脱,检测波长为370 nm,流速为1 mL/min,室温下测定.结果 异鼠李素进样量在0.3~3 μg范围内与峰面积呈良好线性关系(r2=0.999 9),平均回收率为97.93%,RSD为1.55%(n=6),检测限为0.031 2 μg/mL.结论 该方法简单准确、重复性好,适用于五味沙棘颗粒中异鼠李素的含量测定.Objective To establish a high performance liquid chromatography(HPLC) method for the determination of isorhamnetin in Wuwei Shaji Granules.Methods The separation was performed on the Kromsail C18 column (250 mm × 4.6 mm,5 μm) by gradient elution with tetrahydrofuran-acetonitrile-0.4% phosphoric acid aqueous solution(12:18:70) as the mobile phase.The detection wavelength was 370 nm.The flow rate was 1 mL/min.The detecting was performed under ambient temperature.Results The linear range of isorhamnetin was 0.3-3 μg/mL(r2 =0.999 9).The average recovery rate was 97.93%,RSD 1.55% (n =6).The detection limit was 0.031 2 μg/mL.Conclusion This method is simple and accurate with good repeatability,which is suitable for the isorhamnetin determination in Wuwei Shaji Granules.
