流式细胞术检测活化血小板采血方法的改良和护理

摘　　要：目的改良活化血小板流式细胞术检测的采血方法,并探讨其临床应用中的护理配合。方法 2011年3-5月,选择蚌埠医学院第一附属医院健康女性志愿者10例进行采血条件的改良研究,并作为正常对照;选择在蚌埠医学院第一附属医院待产的正常孕妇10例和妊娠期高血压患者10例,用改良的方法检测外周血血小板活化状态。结果 7号和12号针头采集的10份健康志愿者血液标本的活化纤维蛋白原受体(fibrinogen receptor,FIB-R)阳性血小板的百分比和平均荧光强度差异均无统计学意义(均P>0.05);妊娠期高血压患者上述两种指标均明显高于健康非妊娠妇女和正常孕妇(均P<0.01)。结论通过护理人员的配合并注意采血过程中避免人为的血小板活化,可以用7号针头代替12号针头采血,应用于临床流式细胞术血小板活化的检测。Objective To improve the venous blood collection method using in flow cytometric analysis of activated platelets and explore the nursing cooperation methods in its clinical applications.Methods Ten healthy non-pregnant women were selected for the study of improvement of venous blood collection method.In the study,both 7-gauge and 12-gauge needles were used to collect venous blood in same subject for flow cytometric analysis of activated platelets.Ten cases of normal pregnant women and 10 cases of patients with hypertension disorder in pregnancy were selected to analyze the peripheral blood platelet activation using the modified venous blood collection method.Results No significant difference in fibrinogen receptor （FIB-R） positive percentage and its mean fluorescence intensity （MFI） was found between the specimens collected by 7 and 12-gauge needle （P＞0.05）.The FIB-R-positive platelet percentage and MFI in patients with hypertensive disorders in pregnancy were significantly higher than those in healthy non-pregnant women and normal pregnant women （P＜0.01）.Conclusion To avoid artificial platelet activation,the 12-gauge needle can be replaced by the 7-gauge needle in venous blood collection for clinical flow cytometry detection of platelet activation with the collaboration of nursing staff.
