摘 要:目的探讨富半胱氨酸61(cysteine-rich 61protein,cyr61)在哮喘小鼠中的表达与哮喘气道炎症间的关系。方法 24只雌性BALA/c小鼠随机分为哮喘组、地塞米松干预组、孟鲁司特钠干预组及正常对照组,每组6只小鼠。哮喘组通过卵清蛋白(ovalbumin,OVA)致敏和激发的方法制作哮喘小鼠模型。地塞米松干预组在制作哮喘模型同时,于每次激发后给予腹腔注射地塞米松干预。孟鲁司特干预组在制作哮喘模型同时,于每次激发后通过灌胃给予孟鲁司特干预。通过瑞氏-吉姆萨染色分类计数支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中细胞数。通过免疫组织化学技术检测小鼠支气管上皮组织中cyr61的表达。通过ELISA检测BALF中白细胞介素4(interleukin-4,IL-4)和IL-17A含量。结果哮喘组小鼠气道上皮细胞中cyr61蛋白表达明显高于正常对照组小鼠(P<0.05),地塞米松干预组小鼠气道上皮细胞中cyr61蛋白表达与孟鲁司特钠干预组无明显差异,但均较哮喘组明显降低(P<0.05),同时高于正常对照组(P<0.05)。cyr61蛋白表达与BALF中有核细胞总数、嗜酸性粒细胞数、淋巴细胞数、IL-4以及IL-17含量呈正相关(P<0.05)。结论 cyr61蛋白可能参与了哮喘的发病过程,其异常表达可能与气道炎症有关。Objective To explore the expression of cysteine rich 61 protein (cyr61) in asthma mice and the correlation of the expression of cyr61 with airway inflammation in asthma. Methods Twenty-four BALA/c mice were randomly divided into asthma group,dexamethasone group, montelukast group and normal group with 6 mice in each group. Mice were ovalbumin (OVA)-sensitized and challenged to establish asthma models in asthma group,dexamethasone group and montelukast group. In dexamethasone group,dexamethasone was administered intraperitoneally after OVA challenging. Montelukast was administered by gavage in montelukast group. The expression of cyr61 protein in the murine lung tissues was detected by immunohistochemistry. The level of interleukin (IL) 4 and IL-17A protein was determined in the bronchoalveolar lavage fluid (BALF) by enzyme linked immunosorbent assay (ELISA). The number of inflammatory cells in BALF was also counted. Results The ex pression of cyr61 protein was significantly higher in asthma group than that in normal group (P〈0.05). The expression of cyr61 was significantly decreased in both dexamethasone and montelukast groups when compared to asthma group, though it was still higher than that in normal group. The cyr61 protein bore positive correlation to the total nucleated cells (P〈0.05) ,eosinophils (P〈0.05) ,lymphocytes (P〈0.05) ,IL-4 level (P〈0.05) and IL-17A level (P〈0.05) in the BALF. Conclusion Cyr61 may be involved in the pathogenesis of asthma and its abnormal expression may contribute to the airway inflammation in asthma.
