摘  要:目的分析孝感地区RhD阴性个体的分子生物学特征。方法对血清学检测为RhD阴性的标本,通过PCRSSP的方法对RHD特异性外显子7及内含子4,启动子及外显子10进行筛查。阳性标本进一步筛查RHD基因特异性外显子3、4、5、6、7、9及RHD1227A,即DEL。结果在94例RhD阴性标本中,66.0%(62/94)为RHD基因完全缺失,23.4%(22/94)含完整的RHD基因,其中21例为RHD1227A,占22.3%(21/94),10.6%(10/94)为RHD基因部分缺失。结论 RHD基因缺失为孝感地区RhD阴性个体的主要分子生物学特征,DEL为次主要分子生物学特征。Objective To examine the molecular biological characteristics of RhD-negative individuals in Xiaogan area of Hubei province,China. Methods RhD-negative blood samples were obtained and serologically detected. PCR-SSP was used to detect RHD exon 7 and intron 4 in these samples. With samples negative for RHD exon 7 and intron 4, RHD exon 10 and the promoter of the samples were further detected. With samples positive for RHD exon 7 and intron 4, RHD exons 3,4,5,6,7,9 and RHD1227A were determined. Results Complete deletion of RHD gene was found in 62 cases out of all the 94 RhD-negative samples (66%). Complete RHD gene was observed in 22 cases (23.4%) ,among which 21 were RHD1227A (DEL). Ten cases had partial deletion of RHD gene. Conclusion Complete deletion of RHD gene is the main molecular biological characteristics of RhD-negative individuals in Xiaogan area. DEL ranks second in these characteristics.
