摘 要:应用外周微量血培养加细胞松弛素B,观察微核的方法(简称微量CB法),观察^(60)Coγ线低剂量照射诱发的微核和不同年龄组的自发微核,并与常规培养微核法(简称CM法)比较。结果表明外周微量血直接培养,亦获得良好的细胞质分裂阻断微核。两种方法不同年龄组自发微核率,微量CB法的为4.14‰,呈线性正相关。CM法的为6.11‰,随年龄的增长而增多。其低剂量^(60)Coγ线照射诱发的微核,微核率与剂量都有很好的相关性。拟合直线回归方程分别为Y=6.01±0.692D和Y=0.486D-1.968。比较两种方法,CB法更灵敏和准确可靠,可为生物剂量计。This paper reports the cytokinesis-block micronucleus methodin micro-blood cultures. It was performed that CB method in comparison withconventional micronucleus (CM) method for detection induced micronuclei of differentdoses of ^(60)Coγ-rays irradation and spontaneous micronucleus of different ages. The re-sults showed that direct peripheral micro-blood cultures also obtained the cytoknesis-block micronuclei. Using CB method, the micronuclei fequency of different ages wasa linear relationship, Y=1.62+0.74 D, the spontaneous micronuclei frequency of dif-ferent ages was 4.14% the induced. micronuclei also showed that was a linear rela-tionship, Y=6.01+0.692 D. Using CM method, it showed that the induced micronuc-lei was a linear relationship, Y=0.486 D-1.968, but there is no significant differenceseen between the micronuclei frequency of different ages. Comparison with CM anddirect bload smear methods confirmed that the cytokinesis-block method of micro-blo-od cultures is more sensitive and precise.
